The mucopolysaccharides of bovine cornea.

چکیده
The mucopolysaccharides isolated from various mesodermal tissues have, in several instances, been shown to consist of complex mixtures, and not of single compounds as was believed formerly. In view of the rather contradictory findings reported in the literature, it was thought advisable to reinvestigate the mucopolysaccharide fractions of the cornea. In this paper, the separation of three distinct non-dialyzable acid mucopolysaccharides is reported. These, in the order of their precipitation in our fractionation procedure, are (1) a chondroitin sulfate, (2) a uranic acid and hexosamine-containing polymer apparently similar to hyaluronic acid, and (3) a polymer composed of N-acetylglucosamine, galactose, and sulfate in equimolar proportions. This third compound, a novel type of sulfated mucopolysaccharide, we propose to name keratosulfate.

Mucopolysaccharidase of Treponema pallidum.
Treponema pallidum (Nichols strain) exhibited mucopolysaccharidase activity. Acidic mucopolysaccharides were broken down more rapidly by viable treponemes than by heat-inactivated treponemes or membrane filtrates of treponemal suspensions. Ouchterlony immunodiffusion demonstrated the occurrence of antibodies to the hyaluronidase-like enzyme within syphilitic sera. After intratesticular inoculat...
متن کاملMucopolysaccharides in suspensions of Treponema pallidum extracted from infected rabbit testes.
The amount and nature of mucopolysaccharides present in extraction fluids routinely obtained in the isolation procedure of Treponema pallidum from infected rabbit testes was investigated. The mean quantity of mucopolysaccharides extracted from both testes of groups of 10 rabbits was 3.09 mg after infection for seven days and 26.88 mg after infection for 12 days, while from the testes of uninfec...
متن کاملDiagnosis of Hurler's syndrome in the hospital laboratory and the determination of its genetic type.
The rare genetically determined disease now known as Hurler's syndrome, characterized by dwarfism, grotesque skeletal deformity, limitation of joint movement, deafness, hepatosplenomegaly, cardiac abnormality, characteristic facies, and perhaps clouding of the cornea and mental retardation (McKusick, 1960), was first described by Hunter (1917). For some years, the substance that accumulates in ...
متن کاملMucopolysaccharides Produced in Tissue Culture
1. A method of mass tissue culture has been devised by which, in a relatively short period of time, samples large enough for chemical isolation of mucopolysaccharides can be obtained. 2. Chemical isolation of acid mucopolysaccharides from mass cultures of human fetal skin, human fetal bone, bovine fetal skin, and rat subcutaneous tissue has been carried out. It has been found that the fibroblas...
متن کاملGlycosaminoglycans in the Human Cornea: Age-Related Changes
AIM To investigate possible age-related changes in glycosaminoglycans (GAGs) in the human cornea. The substances today called GAGs were previously referred to as mucopolysaccharides. METHODS Samples of human cornea were taken from 12 younger (age 21 ± 1.2) and 12 older (age 72 ± 1.6) male subjects. Samples were weighed, homogenized, and used for biochemical and molecular analyses. All the qua...
