Postischemic Reperfusion Injury

چکیده
We studied the role of copper as a potential mediator of postischemic reperfusion injury in the isolated, perfused rat heart. Hearts were equilibrated with Krebs-Henseleit buffer for 10 minutes and then loaded with copper by way of perfusion with buffer containing 20 pM copper(II)-bis-histidial for 30 minutes. Control hearts were perfused with Krebs-Henseleit buffer alone during the loading period. Hearts then were washed with buffer for 10 minutes and subjected to 20 minutes of normothermic global ischemia followed by 30 minutes of reperfusion. Atomic absorption spectroscopy revealed a 67% increase in total copper content in loaded hearts by the end of the wash. By the end of the 30-minute period of reperfusion, control hearts demonstrated a 50-60% recovery of myocardial function as determined by peak systolic

Effect of endogenous nitric oxide on cardiac ischemic preconditioning in rat
Introduction: Ischemic Preconditioning (IPC) is the phenomen that happens on the heart by one or several short periods of ischemia followed by reperfusion that improve the postischemic recovery of mechanical function. Ischemic preconditioning (IPC) may protect the heart from ischemia reperfusion injury by nitric oxide formation. This study investigated the effect of ischemic preconditioni...
متن کاملGentamicin effects on renal ischemia/reperfusion injury.
This study assessed gentamicin's effects on ischemia/reperfusion renal injury to better understand when and how it worsens postischemic acute renal failure. Rats were subjected to 25 minutes of renal pedicle occlusion with and without preischemic (15-minute) or postischemic (15-minute or 8-hour) gentamicin treatment (100 mg/kg, by itself a subtoxic dose). Gentamicin's impact on hypoxia/reoxygen...

متن کاملAmelioration of postischemic reperfusion injury by antiarrhythmic drugs in isolated perfused rat lung.
Antiarrhythmic drugs, such as lidocaine, quinidine, and procainamide, have been shown to be effective against postischemic reperfusion injury in isolated rat lungs. Rat lungs were perfused at a constant flow with Krebs-Henseilet buffer supplemented with 4% bovine serum albumin and ventilated with air containing 5% CO2. The lungs were subjected to ischemia by stopping perfusion and ventilation f...
متن کاملGlutathione treatment protects the rat liver against injury after warm ischemia and Kupffer cell activation.
BACKGROUND/AIM The generation of reactive oxygen species by activated Kupffer cells (KC) may contribute to reperfusion injury of the liver during liver transplantation or resection. The aim of our present studies was to investigate (1) prevention of hepatic reperfusion injury after warm ischemia by administration of the antioxidant glutathione (GSH) and (2) whether GSH confers protection throug...
