Rohon-Beard neuron origin from blastomeres of the 16-cell frog embryo.

چکیده

Clonal origins of Rohon-Beard neurons in Xenopus were determined quantitatively by injecting horseradish peroxidase into individual blastomeres at the 16-cell stage and later counting labeled and unlabeled Rohon-Beard neurons. Two different patterns of cleavage were selected. In pattern X, all Rohon-Beard neurons originated from three blastomeres (V1.1, V1.2, and V2.2) on each side; in pattern Y, all Rohon Beard neurons originated from two blastomeres (V1.2 and V2.2) on each side. Counts of Rohon-Beard neurons at larval stages 32 to 34 showed that 96 to 100% (mean 99%) originated from blastomeres on the same side; of these, 68 to 90% (mean 75%) descended from V1.2, 20 to 31% (mean 24%) descended from V2.2, 0 to 7% descended from V1.1. The significance of the regionally restricted origin of Rohon-Beard neurons is discussed.

Rohon-Beard neurons arise from a substitute ancestral cell after removal of the cell from which they normally arise in the 16-cell frog embryo.

In 16-cell Xenopus embryos, horseradish peroxidase (HRP) was injected into blastomere D1.2 on one side. No Rohon-Beard neurons originated from D1.2 in either of the two patterns of cleavage that were studied (Jacobson, M. (1981) J. Neurosci. 1: 918-922). In other embryos, after injection of HRP into D1.2, the neighboring ventral blastomere V1.2, from which 68 to 90% of Rohon-Beard neurons norma...

Quantitative lineage analysis of the frog's nervous system. I. Lineages of Rohon-Beard neurons and primary motoneurons.

After injection of horseradish peroxidase into single blastomeres in Xenopus embryos at 2- to 512-cell stages, all of the descendants could be traced and counted at tailbud stages. Progenitors of Rohon-Beard neurons and of primary spinal motoneurons were identified, and all neurons of each type that originated from individual progenitors were counted. From these data were derived the geometric ...

Skin impulse excitation of spinal sensory neurons in developing Xenopus laevis (Daudin) tadpoles.

Responses to gentle touch in young Xenopus tadpoles are mediated by spinal cord sensory Rohon-Beard neurons. Tadpoles also respond to noxious stimuli that elicit 'skin impulses', which propagate between epithelial cells over the whole body surface, somehow entering the CNS to generate a response. After hatching (~48 h post-fertilization), skin impulse signals enter the CNS only via cranial nerv...

Delta/Notch signaling promotes formation of zebrafish neural crest by repressing Neurogenin 1 function.

In zebrafish, cells at the lateral edge of the neural plate become Rohon-Beard primary sensory neurons or neural crest. Delta/Notch signaling is required for neural crest formation. ngn1 is expressed in primary neurons; inhibiting Ngn1 activity prevents Rohon-Beard cell formation but not formation of other primary neurons. Reducing Ngn1 activity in embryos lacking Delta/Notch signaling restores...

I-38: Chromosome Instability in The Cleavage Stage Embryo

Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...
