Gentiobiose, a constituent of deoxyribonucleic acid from coliphage T6.

چکیده
The deoxyribonucleic acid of the T-even coliphages contains glucosylated and nonglucosylated 5-hydroxymethylcytosine in place of cytosine (l-6). In the case of deoxyribonucleic acid from phage T6, 72y0 of the 5-hydroxymethylcytosine residues are diglucosylated. In T2 deoxyribonucleic acid, 70% of the 5-hydroxymethylcytosine residues are monoglucosylated, but 5% bear diglucosyl units. A recent investigation of the configuration of the disaccharides bound to the 5-hydroxymethylcytosine nucleotides of deoxyribonucleic acid from T2 and T6 showed the glucose units to be p linked to each other and joined to the hydroxymethyl group of the 5-hydroxymethylcytosine by an cr linkage (6). In the present study, the disaccharide from T6 deoxyribonucleic acid was identified as gentiobiose,

The deoxyribonucleic acid of the bacterial viruses, T2, T4, and T6, is unique in containing the pyrimidine base, 5-hydroxymethylcytosine, in place of cytosine (2). Attached to a fraction of the 5-hydroxymethylcytosine residues are glucosyl or diglucosyl units in amounts and linkages characteristic for each of the phages. 5-Hydroxymethylcytosine residues with ar-monoglucosyl substituents occur i...
متن کاملGlucosylation of Deoxyribonucleic Acid by Enzymes
The deoxyribonucleic acid of bacteriophages T2, T4, and T6 contains glucose linked to the hydroxymethyl groups of hydroxymethylcytosine. The work of Sinsheimer, Volkin, Jesaitis, and of Lichtenstein and Cohen (l-4) established the presence of glucosyl-hydroxymethylcytosine residues in the deoxyribonucleic acid and indicated that there is a characteristic glucose to hydroxymethylcytosine ratio f...
متن کاملSeparation of T-even bacteriophage deoxyribonucleic acid from host deoxyribonucleic acid by hydroxyapatite chromatography.
A simple and rapid method is described for separation of T-even bacteriophage deoxyribonucleic acid (DNA) from host (Escherichia coli) DNA by hydroxyapatite column chromatography with a shallow gradient of phosphate buffer at neutral pH. By this method, bacteriophage T2, T4, and T6 DNA (but not T5, T7, or lambda DNA) could be separated from host E. coli DNA. It was found that glucosylation of t...

Introduction: Low-energy helium-neon (He-Ne) laser beam lightis used in combination with sodium hypochlorite (Na2HOCl3) for clinical purposes. Regarding this, the present study aimed to investigate the effect of He-Ne laser (632.8 nm) and sodium hypochlorite on the calf thymus double-stranded deoxyribonucleic acid (ctdsDNA) molecule. Materials and Methods: For the purpose of the study, ctdsDNA...
متن کاملGlucosylated Nucleotide Sequences from T-even Bacteriophage Deoxyribonucleic Acids.
1. The frequencies of various pyrimidine nucleotide sequences in phage-T2VG111 DNA and phage-T6 DNA have been measured. The hydroxymethylcytosine nucleotides that bear glucosyl groups in phage-T2VG111 DNA and gentiobiosyl groups in phage-T6 DNA are not randomly distributed. 2. Sequences in which two or three hydroxymethylcytosine nucleotides are terminated at both ends by purine nucleotides bea...
