Initial Drug Substance Characterization (IDSC) of a novel anti – white spot syndrome virus drug derived from terrestrial plants

چکیده
White spot syndrome virus (WSSV), the most contagious pathogen of cultured shrimp, causes mass mortality, leading to huge economic loss to the shrimp industry. The lack of effective therapeutic or prophylactic measures has aggravated the situation, necessitating the development of antiviral drugs. With this objective, the antiviral activity of the drug, (TP22C derived from the terrestrial plant) in Litopenaeus vannamei was evaluated. The Initial Drug Substance Characterization (IDSC) such as, physical characterization, structural elucidation and preformulation data analysis of TP22C was carried out. The in vivo analysis of TP22C in the host – pathogen interaction model revealed the efficiency of the drug. The survival percentage of the treated (with TP22C) WSSV infected host was 86 %. The 10 phytomolecules, present in the TP22C was illustrated by these analytical tools. The druggability of these molecules was determined by the techniques of computational chemistry and chemometrics. This overall assessment proved the phytomolecular consortium (TP22C), to be a potent drug and can protect L. vannamei against the WSSV infection. © 2013 Universal Research Publications. White spot syndrome virus (WSSV) is a highly pathogenic and prevalent virus affecting shrimp culture worldwide including Iran. In the present study, a pair of primers was designed according to the sequence of VP 28 gene of WSSV in the GenBank. VP28 gene from an Iranian WSSV isolate (IrVP28) was cloned, sequenced and expressed in Escherichia coli BL21(DE3) strain in order to produce VP28 protein...
