چکیده
Human papillomaviruses (HIPVs), especially types 16 and 18, are strongly associated with the development of cervical intraepithelial neo. plasia(CIN)and Invasivecarcinoma. The HPV E6 and E7 proteinsare expressed constitutively in the majority of CIN lesions and carcinomas. Therefore, they are targets for the inunune response against HPV and candidates for active immunotherapy. We have previously detected HPV specific CTLs from the peripheral blood mononuclear cells from a cervi cal cancer patient following immunization with a recombinant vaccinia using in vitro restimulation with adenovirus recombinants expressing HPV16 or HPV1S E6/E7 fusion proteins. In this study, we used a similar protocol to determine the prevalence of CTL responses against HPV16 andHPV18E6/E7Intheperipheralbloodmononuclearcellsfrom10CIN Ill and 10normalsubjects.HPV-speclflcCTLresponsesweredetectedin 6 of 10 CIN III subjects. These CTL lines recognized HPV16 E61E7 proteins presented by at least three MHC class 1 HLA alleles and by HPV-transformedCaSkicells. No HPV-speciflcCTLs were detectedIn normal subjects. This study demonstrates the presence of naturally oc curling HPV-speclflc memory CTLs In a majority of CIN III patIents and provides an approach for further study of their role in modulating cer vical malignancy.

Activation Breast Cancer: Characterization and Antigen-specific among Tumor-infiltrating Lymphocytes from a Patient with MAGE-1-specific Precursor Cytotoxic T-Lymphocytes Present
A potential target for development of tumor-specffic immunotherapeu tic strategies is the MAGE-1 gene. We have utilized a recently developed recombinant canarypox (ALVAC) virus vector containing the MAGE-1 gene (vCP23S) to activate CTLs from a breast cancer patient bearing a MAGE-1@tumor. Tumor-infiltrating lymphocytes (TILs) obtained from the tumor of a patient were stimulated in vitrowith irr...

A potential target for development of tumor-specific immunotherapeutic strategies is the MAGE-1 gene. We have utilized a recently developed recombinant canarypox (ALVAC) virus vector containing the MAGE-1 gene (vCP235) to activate CTLs from a breast cancer patient bearing a MAGE-1+ tumor. Tumor-infiltrating lymphocytes (TILs) obtained from the tumor of a patient were stimulated in vitro with ir...
متن کاملImmunization with human papillomavirus type 16 (HPV16) oncoprotein-loaded dendritic cells as well as protein in adjuvant induces MHC class I-restricted protection to HPV16-induced tumor cells.
Human papillomavirus (HPV) E6 and E7 oncoproteins are attractive targets for T-cell-based immunotherapy of cervical cancer. In this study, we demonstrate that dendritic cells (DCs) pulsed with HPV16 E7 protein are not only recognized in vitro by E7-specific CTLs but also elicit E7-specific CTL responses in vivo, associated with protection against a challenge with syngeneic HPV16-induced tumor c...
متن کاملTherapeutic and anti-metastatic effect of CIKs on cervical cancer enhanced by modified HPV16 E7-pulsed DCs with SOCS1-knockdown
Cervical cancer is the second most frequent cancer and the second most common cause of cancerrelated deaths among women worldwide. In this study, a modified HPV16 E7 (HPV16mE7) with reduced transformation activity and enhanced antigenicity was employed to load DCs with silenced suppressor of cytokine signaling 1 (SOCS1) gene. The prepared DCs were co-cultured with peripheral blood mononuclear c...
متن کاملEvaluation of in vitro Effect of Pyocyanine Pigment on Interleukin-2 Secretion from Peripheral Blood Mononuclear Cells in Cancer Patients
Background and Objective: The pyocyanine pigment in Pseudomonas aeruginosa stimulates blood cells to secrete IL-2. IL-2 cytokine is an activator of cytotoxic T cells and natural killer cells. These cells destroy the target cells of patients with cancer. This study aimed to evaluate the effects of pyocyanine on the IL-2 secretion from peripheral blood mononuclear cells (PBMCs) in patients with b...
