چکیده

Since the discovery of vitamin Bs, several biological assay procedures for this factor have been proposed (Gyorgy (1) ; Lunde and Kringstad (2); Wilson and Roy (3) ; Schneider et al. (4) ; Halliday and Evans (5) ; Edgar, El Sadr, and Macrae (6) ; Dimick and Schreffler (7); Reedman et al. (8)). These methods, with the exception of that of Edgar et al., are based upon the cure of acrodynia produced on a vitamin Bs-deficient ration or upon a combination of growth and the cure of acrodynia of vitamin Bedepleted rats. Edgar et al. propose an assay based on the growth response alone. In light of the fact that factors other than vitamin Be are concerned in the production and alleviation of dermatitis in the rat, methods involving the curing of dermatitis present difficulties in technique and interpretation (Birch (9) ; Schneider et al. (10)). Furthermore some workers have not been able to produce the dermatitis consistently (Dann (11)). The method described here is based entirely upon the growth response of vitamin Be-depleted rats supplemented either with synthetic pyridoxine or the substance to be assayed. Two rations are described. In Ration I a fullers’ earth filtrate of a butanol extract of 1: 20 liver concentrate powder is used to supply pantothenic acid and unidentified factors necessary for the rat. When synthetic pantothenic acid became available, Ration II was devised in which synthetic pantothenic acid and 0.25 per cent of

THE VITAMIN Ba GROUP VIII. BIOLOGICAL ASSAY OF PYRIDOXAL, PYRIDOXAMINE, AND PYRIDOXINE*

Though various chemical and microbiological methods have been developed for the estimation of vitamin Bs in whole (2-4) or in part (5), animal assays are still necessary for testing the accuracy of these new methods as they apply to animal nutrition. It is also important that the relative potency of pyridoxine, pyridoxamine, and pyridoxal for animals be known. Two different purified rations hav...

Identification of pyridoxine 3-sulfate, pyridoxal 3-sulfate, and N-methylpyridoxine as major urinary metabolites of vitamin B6 in domestic cats.

Preliminary studies of vitamin B6 metabolism in three adult domestic cats detected very little pyridoxic acid in the urine. At oral doses of 49 to 490 mumol of [14C]pyridoxine hydrochloride, 50% of the excreted dose occurred as pyridoxine 3-sulfate and 25% as N-methylpyridoxine. The identity of these two metabolites was confirmed by isolation from urine and comparison with known compounds. A th...

The determination of 4-pyridoxic acid in human urine.

The existence of a fluorescent compound in human urine after the ingestion of vitamin Be was reported by Singal and Sydenstricker (1) in 1941. Huff and Perlzweig (2, 3) isolated a pyridosine metabolite from human urine which they identified as 2-methyl-3-hydroxy-4-carboxy-5-hydroxymethylpyridine (Cpyridoxic acid). A fluorometric method for the determination of this substance was also described ...

Influence of dietary pyridoxine on glutamic decarboxylase activity of brain.

y-Aminobutyric acid is present in the free form in large amounts in brain (l-3), where it is formed from glutamic acid by a L-glutamic acid decarboxylase (2, 4, 5). The enzyme requires pyridoxal phosphate as coenzyme (5), which can be synthesized in brain from adenosinetriphosphate and pyridoxal (6). This decarboxylase has a high degree of substrate and coenzyme specificity (6). The concentrati...

An effect of pyridoxine on blood urea in human subjects.

In 1942 pyridoxine was related to protein metabolism (1). Since that time this vitamin or a derivative thereof has been described as a component of transaminase (2) and of decarboxylase (3). Pyridoxine has been claimed to be beneficial in the treatment of a variety of diseases. An attempt to produce a deficiency of this vitamin in a human subject failed to give evidence of any significant chang...
