Characterization of the matK Gene in the Poaceae

چکیده
The matK gene is named according to its possible maturase function and its location within the trnK gene encoding the tRNA (UUU). A 509 open reading frame was found to reside in an intron when the trnK gene was sequenced in tobacco (Nicotiana tabacum) (Sugita et al., 1985). Later, a complete sequence of the liverwort Marchantia polymorpha confirmed the existence of this open reading frame in the non-vascular plant, although it is short compared to those in other plants (Ohyama et al., 1986). Neuhaus and Link (1987) suggested the possible maturase function of the matK gene in mustard (Sinapis alba), based upon their homology search result. A segment near the carboxyl terminus of the derived mustard trnK polypeptide is structurally related to the portions of maturase-like polypeptides of introns of the mitochordrial cytochrome c oxidase subunit I gene (COXI) of yeast (Saccharomyces cerevisiae) and ascomycete (Podospora anserina). The maturase MatK presumably helps fold the intron RNA into the catalytically-active structure. Thereafter, the matK gene was sequenced from various plants, such as rice (Oryza sativa), two pine species (Pinus contrta and P. thunbergii), and beechdrop (Epifagus virginiana) (Hiratsuka et al., 1989; Lidholm and Gustafsson, 1991; Wolfe et al., 1992). In beechdrop, this open frame appeared as a freestanding gene with neither the trnK exons nor the interrupting introns present.

Cloning and molecular characterization of TaERF6, a gene encoding a bread wheat ethylene response factor
Ethylene response factor proteins are important for regulating gene expression under different stresses. Different isoforms for ERF have previously isolated from bread wheat (Triticum aestivum L.) and related genera and called from TaERF1 to TaERF5. We isolated, cloned and molecular characterized a novel one based on TdERF1, an isoform in durum wheat (Tri...
متن کاملAssessing universality of DNA barcoding in geographically isolated selected desert medicinal species of Fabaceae and Poaceae
In pursuit of developing fast and accurate species-level molecular identification methods, we tested six DNA barcodes, namely ITS2, matK, rbcLa, ITS2+matK, ITS2+rbcLa, matK+rbcLa and ITS2+matK+rbcLa, for their capacity to identify frequently consumed but geographically isolated medicinal species of Fabaceae and Poaceae indigenous to the desert of Cholistan. Data were analysed by BLASTn sequence...
متن کاملMolecular Phylogeny of the Genus Lathyrus (Fabaceae-Fabeae) Based on cpDNA matK Sequence in Iran
Background: More than 60 species of the genus Lathyrus are distributed in Southwest Asia. It is the second largest genus of the tribe Fabeae, after Vicia, in the region (and in Iran with 23 species). In the regional Flora (Flora of Turkey, FloraIranicaand flora...
متن کاملThe matK gene: sequence variation and application in plant systematics.
Although the matK gene has been used in addressing systematic questions in four families, its potential application to plant systematics above the family level has not been investigated. This paper examines the rates, patterns, and types of nucleotide substitutions in the gene and addresses its utility in constructing phylogenies above the family level. Eleven complete sequences from the GenBan...
متن کاملEvolutionary Implications of Matk Indels in Poaceae1
Insertion/deletion events (indels) and nucleotide substitutions at the extreme 39 end of the chloroplast gene matK have been identified that distinguish certain major lineages of grasses. A 1-bp (base pair) deletion creating a shift in the open reading frame (ORF) and a point mutation support the positions of Streptochaeta and Anomochloa as the two most basal lineages in Poaceae. Another 1-bp d...
