Viscum album L. and Quercetin Reduce Cyclophosphamide-Induced Toxicity in Mice

چکیده
Cyclophosphamide (CP) is an alkylating agent and its tumor cell-killing activity is mainly due to its DNA alkylation. Phosphoramide mustard and acrolein are the two active metabolites of CP. CP metabolites can react with carboxyl (-C[O]OH), mercapto (-SH), amino (-NH2), phosphate (-PO3H2) and hydroxyl (-OH) groups, and can form cross-links with DNA and proteins (Todorova et al., 2009). The precise mechanism by which CP causes toxicity is unknown; however numerous studies have shown that CP exposure can disrupt the redox balance of tissues, suggesting that biochemical and physiological disturbances may result from oxidative stress. Accordingly, the toxic effects of CP on cardiac tissues (Mythili et al., 2005; Senthilkumar et al., 2006; Todorova et al., 2009; Motawi et al., 2010; Nagi et al., 2010) and hemorrhagic cystitis (Bhatia et al., 2006; Linares-Fernández and Alfieri, 2007; Bhatia et al., 2008; Arafa, 2009; Motawi et al., 2010; Santos et al., 2010) were demonstrated in different animal models. Furthermore, in somatic cells, CP has been shown to produce gene mutations, DNA-strand

Screening of Neuropharmacological Activities of Viscum album and Estimation of Major Flavonoid Constituents Using TLC Densitometry
Despite a long tradition of use in the treatment of mental disorders, Viscum album L. (Ujjral; family – Viscaceae) has not been systematically investigated to validate its traditional claims. The methanol extract of V. album aerial parts, its ethyl acetate fraction (EAF) and 1-butanol fraction (BF) were screened for various neuropharmacological activities. The methanol extract, EAF and BF exhib...
متن کاملAnti-inflammatory and antinociceptive activity of flavonoids isolated from Viscum album ssp. album.
Viscum album L. has been used in the indigenous systems of medicine for treatment of headache and some inflammatory diseases. In order to evaluate this information, antinociceptive and anti-inflammatory activities of the five flavonoids (5,7-dimethoxy naringenin or 4',6'-dimethoxy chalcononaringenin) derivatives, isolated from the ethyl acetate fraction of the extract from V. album ssp. album, ...

Ethanolic and aqueous extracts from five plant species used in Turkish traditional medicine were evaluated for in vivo hypercholesterolaemic and antioxidant activities: Agrostemma githago L., Potentilla reptans L., Thymbra spicata var. spicata L., Urtica dioica L. and Viscum album var. album L. We assayed the effects of the administration of plant extracts on serum total cholesterol, triglyceri...
متن کاملThe Protective Effects of Origanum vulgare L. Extract on Genetic Damage of Cyclophosphamide in Mice Blood Lymphocytes Using Micronucleus Test
Despite its various clinical applications, cyclophosphamide (CP), an alkylating chemotherapeutic agent, has demonstrated numerous side effects, including genetic toxicity. This study investigated the protective action of Origanum vulgare L., a powerful antioxidant plant, on the genotoxicity of CP in the mice blood lymphocytes. The mice were pretreated orally with different doses of (50, 100, 20...

Additional therapy with extracts of Viscum album [L.] (VaL) increases the quality of life of patients suffering from early stage breast cancer during chemotherapy. In the current study patients received chemotherapy, consisting of six cycles of cyclophosphamide, anthracycline, and 5-Fluoro-Uracil (CAF). Two groups also received one of two VaL extracts differing in their preparation as subcutane...
