Inhibition of nitric oxide synthase accentuates endotoxin-induced sickness behavior in mice

چکیده
Sickness behavior appears to be the expression of a central motivational state that reorganizes an organism's priorities to cope with infectious pathogens. To evaluate the possible participation of nitric oxide (NO) in lipopolysaccharide-induced sickness behaviors, mice were submitted to the forced swim test (FST), open field test and dark-light box test. Food intake and corticosterone plasma levels were evaluated. Lipopolysaccharide (LPS, 100 μg/kg; i.p.) administration increased the time spent floating in the FST and decreased locomotor activity in the open field. Indeed, treatment with LPS decreased the total number of transitions between the dark and light compartments of the apparatus. In addition, LPS reduced food intake and increased corticosterone levels. Pretreatment with L-NAME (30 mg/kg; i.p.) or aminoguanidine (50mg/kg; i.p.) accentuated the behavioral changes induced by LPS in the FST, open field and light-dark box tests as well as induced an increment in hypophagia and in corticosterone levels. These findings confirm previous observations that have reported LPS-induced sickness behaviors. In addition, they provide evidence that the synthesis of NO modulates changes in depressive-like and exploratory behaviors in mice, which is supported by the fact that NO synthase inhibitors also attenuate LPS-induced behavioral changes. In addition, the present study suggests that NO may have a protective role, acting in an inhibitory feedback manner to limit LPS-induced sickness behavior.

Inhibition of nitric oxide synthase activity improves focal cerebral damage induced by cerebral ischemia/reperfusion in normotensive rats
Introduction: Nitric oxide seems to play a dual role in ischemia/reperfusion injury. Few studies have investigated whether it exacerbates or improves brain edema. In the present study, we inhibited the activity of nitric oxide synthase by L-NAME and evaluated the cerebral infarct volume, tissue swelling and brain edema, alongside the measurement of blood flow of the ischemic region. Methods...
متن کاملCongenital deficiency of nitric oxide synthase 2 protects against endotoxin-induced myocardial dysfunction in mice.
BACKGROUND Sepsis can be complicated by severe myocardial dysfunction and is associated with increased nitric oxide (NO) production by inducible NO synthase (NOS2). To investigate the role of NOS2 in endotoxin-induced myocardial dysfunction in vivo, we studied wild-type and NOS2-deficient mice. METHODS AND RESULTS Serial echocardiographic parameters of myocardial function were measured before...
متن کاملRole of matrix metalloproteinase II on analgesic effect of nitric oxide inhibition in rat
Abstract Introduction: Matrix metalloproteinase 2 is one of the inflammatory mediators that is involved in nociceptive processing and its production is regulated by many inflammatory factors such as nitric oxide. We studied the role of MMP-2 on the analgesic effects of nNOS inhibitor. Methods: Considering that nitric oxide has many roles in pain processing, we studied the CSF levels of MMP-2 ...
متن کاملA Selective Inducible Nitric Oxide Synthase Dimerization Inhibitor Prevents Systemic, Cardiac, and Pulmonary Hemodynamic Dysfunction in Endotoxemic Mice
Increased nitric oxide (NO) production by inducible NO synthase (NOS2), an obligate homodimer, is implicated in the cardiovascular sequelae of sepsis. We tested the ability of a highly-selective NOS2 dimerization inhibitor (BBS-2) to prevent endotoxin-induced systemic hypotension, myocardial dysfunction, and impaired hypoxic pulmonary vasoconstriction (HPV) in mice. Mice were challenged with Es...
متن کاملEvaluation of nitric oxide involvement in effect of lead on dependency to morphine in mice
In the present study, interactions between lead exposure with nitric oxide precursor (L-arginine) or nitric oxide synthase (NOS) inhibitor (L-NAME) on naloxone-induced jumping and diarrhea in morphine-dependent mice were examined. Chronic lead acetate (0.05%) exposure altered naloxone-induced jumping and diarrhea in mice. Jumping was decreased after 7 days and was unchanged 14 and 28 days after...
