چکیده

It is now possible to store turkey semen for 4 to 24 h without appreciable loss in fertility and hatchability. Optimum conditions for 24 or 48 h storage have not been fully determined. Development of new storage media, allowing the insemination of very low numbers of spermatozoa, may provide interesting possibilities regarding the use of superior sires. This study was conducted to evaluate effects of simple sugar-based extenders (0.3 M Glucose, 0.6 M Sucrose) or Ringer + glucose solution after 24 h or 48 h of cooled storage on fertility. Standard bred bronze turkey toms 40 weeks of age were used. Semen was collected by abdominal massage twice a week throughout 4 weeks of study. Ejaculates were pooled, mixed thoroughly, and diluted to insemination dose (200 x 10) with 1) 0.3 M glucose, 2) 0.6 M sucrose, and 3) ringer + glucose solution. Diluted semen were placed in 10 ml erlenmeyer flasks. All flasks were covered with parafilm and stored in a refrigerator for 24 or 48 h at 4 C. Changes in sperm motility were determined before and after holding. Sperm stored in 0.6 M sucrose solution was not used for the inseminations because of low motility (10 % after 24 h, 0 % after 48 h). Hens were inseminated two times in the first week and once weekly with 200 x 10 viable spermatozoa for 4 weeks. The mean fertility rates for sperm stored with two extenders (0.3 M glucose and Ringer + glucose) at 4 C were: 90.9% ( 24 h, 0.3 M glucose); 79.1% ( 48 h, 0.3 M glucose); 69.8 % (24 h, Ringer + glucose); 30.9 % (48 h, Ringer + glucose); 93.8 % (control, fresh). No significant difference in fertility rates were observed between sperm stored in glucose-based extender for 24 hr and the unstored control, but differences between extenders were significant (p<0.05). Thus, glucose-based extender was beneficial and provided better conditions for cooled storage than did the others. There was no difference in hatchability of fertile eggs in the treatments. The results show that turkey semen can be stored in glucose-based extender for 24 or 48 h with a minimal loss in fertility and hatchability.

Fertilizing ability of short-term preserved spermatozoa Abant trout (Salmo trutta abanticus

The objective of this experiment was to evaluate spermatological parameters and fertilizing ability of short term stored semen in different extenders from endemic Abant trout (Salmo trutta abanticus T,1954). Semen was collected from 30 adult males by the hand stripping method without anesthesia. Having determined the main spermatological properties (volume, motility, movement duration, concentr...

Maintenance of ATP concentrations in and of fertilizing ability of fowl and turkey spermatozoa in vitro.

Fowl and turkey spermatozoa, when diluted in a glutamate-based medium and incubated at 40\s=deg\Cunder aerobic conditions showed a similar rate of oxygen utilization and maintained similar cellular ATP concentrations. Under anaerobic conditions, the glycolytic metabolism of fowl spermatozoa was sufficient to maintain a high ATP concentration, whilst turkey spermatozoa had a much lower glycolyti...

Effects of Extender and Storage Time on Motility and Ultrastructure of Cooled-Preserved Boar Spermatozoa

Artificial insemination (AI) using fresh diluted semen is used worldwide in the porcine industry. Viability of spermatozoa in diluted semen depends on several factors, such as the interaction with the type of extender and storage duration. The aim of this study was to evaluate the effects of extenders and storage time on the motility and ultrastructure of cooled-preserved boar spermatozoa. Seme...

Fertilizing ability of cryopreserved spermatozoa in the Persian sturgeon (Acipenser persicus) and stellate sturgeon (A. stellatus)

Motility of spermatozoa was studied on 12 and 7 specimens of Acipenser persicus and A. stellatus, respectively. The density measured to be 2.22±0.65x109ml-1 in A. persicus and 2.21±0.55x109ml-1 in A. stellatus. Semen samples were diluted with two extenders containing tris 118mM, sucrose 23.4mM, pH=8, egg yolk (20%), dimethyl sulfoxide (15%) and penicillin potassium (500IU/ml) and biociphus exte...

The effects of methylxanthines on the mobility of stored turkey sperm.

The mobility of pooled turkey sperm following various storage regimens was assessed by objectively measuring the ability of sperm to penetrate a 2% Accudenz [5-(N-2,3-dihydroxypropylacetamido)-2,4,6-tri-iodo-N,N'-bis(2,3-dihydroxypropyl)isophthalamide] solution at 41 C. When semen was diluted with Beltsville poultry semen extender and stored at 5 C with agitation at 150 rpm, sperm mobility decl...
