چکیده

The present study was conducted to investigate the existence of polymorphism at Kappa-casein (κ-CN) locus in the Nili-Ravi buffalo. A PCR-RFLP method was used to distinguish buffalo κ-CN alleles. The genotypes were confirmed through specific, clearly distinguishable DNA band pattern resulting from digestion with three selected restriction enzymes (HinfI, HaeIII and MaeII). Analysis of 163 animals revealed that all animals were monomorphic, showing only BB genotype. It was concluded that κ-CN B allele, which has significant effect on milk quality reported previously, may be included in breeding strategies of dairy animals. The monomorphism for κ-CN locus observed in buffalo population can lead to the development of a method to identify mixtures of cow and buffalo milk in cheese processing.

First Report on Standard G-Banded Karyotype of Nili-Ravi Buffalo, Bubalas bubalis

The present study was planned to determine if chromosomal dysmorphology based on G band nomenclature is common in the Nili-Ravi buffalo which is the prime dairy breed thriving predominantly in the Punjab Province of Pakistan. Microscopic screening of 50 good quality metaphase cells from each of 34 male and female buffalo calves revealed the expected chromosome complement of 24 autosomal pairs a...

Genetic diversity analysis of an indigenous Chinese buffalo breed and hybrids based on microsatellite data.

Chinese native buffaloes have faced the threat of extinction, along with an increase in crossbreeding with domesticated river buffaloes; consequently, conservation of local buffalo genetic resources has become a priority. A Chinese native breed, Jianghan, is often crossed intentionally and unintentionally with imported breeds from India and Pakistan, Murrah, and Nili-Ravi. A total of 128 b...

Genetic polymorphism and association of kappa-casein gene with milk production traits among Frieswal (HF × Sahiwal) cross breed of Indian origin

The aim of the present study was to screen the genotype profile of bovine kappa-casein gene among Frieswal (HF × Sahiwal) crossbred cattle developed in India. A total number of two hundred Frieswal cows were evaluated for HinfI RFLP based genotyping of kappa-casein gene. We observed that only two genotypes (AA and AB) exist among the studied population with the genotype frequency of 0.58 (n=117...

Investigation of Kappa Casein Gene Polymorphism by PCR-RFLP in Najdi Cattle Breed Population in Khuzestan Province

More than 80% of the total milk protein contains caseins that are to forms α-s1, α-s2, β-casein and kappa casein. Kappa casein is smaller than other milk proteins but due to have a role in size and stability of micelles. The present study describes polymorphism of kappa casein gene. This is the first study of kappa casein gene polymorphism in Najdi cattle of Iran. We used the polymerase chain r...

Characterization of Indian Riverine Buffaloes by Microsatellite Markers

Six breeds of riverine buffalo viz. Murrah, Mehsana, Jaffrabadi, Nagpuri, Nili-Ravi and Bhadawari were characterized using FAO-recommended cattle specific microsatellite markers. Among the total of twenty microsatellite markers screened to explore genomic variability of six buffalo breeds, only ten were polymorphic in nature. Four out of ten polymorphic microsatellite loci were rated as informa...
