چکیده

Purpose: To investigate the inhibitory effects of various ratios of polysaccharides/ alkaloids from the rhizome of Coptis chinensis Franch (RCC) on α-glucosidase. Methods: The polysaccharides (PSD) and alkaloids (ALK) from RCC were prepared using the water extraction and alcohol precipitation method and Reinecke’s salt precipitation method, respectively. Subsequently, the α-glucosidase inhibitory effects of PSD, ALK, and PSD/ALK at various ratios were evaluated spectrophotometrically in vitro. Results: With a half maximal inhibitory concentration (IC50) value of 171.67 μg/mL, ALK showed higher α-glucosidase inhibitory activity than PSD (IC50 = 296.89 μg/mL). In addition, the polysaccharides/alkaloids (PSD/ALK) at the ratio of 3:1 exhibited stronger α-glucosidase inhibitory activity (IC50 = 160.9 μg/mL) than PSD, ALK and PSD/ALK at ratios of 1:3 (IC50 = 394.78 μg/mL), 1:2 (IC50 = 185.18 μg/mL), 1:1 (IC50 = 350.51 μg/mL), and 2:1 (IC50 = 229.16 μg/mL). Conclusion: The results obtained suggest that PSD/ALK (3:1) possesses the strongest α-glucosidase inhibitory effect and may be considered as a candidate agent in future anti-diabetes drug development.

Transcriptome analyses provide insights into the difference of alkaloids biosynthesis in the Chinese goldthread (Coptis chinensis Franch.) from different biotopes

Coptis chinensis Franch., the Chinese goldthread ('Weilian' in Chinese), one of the most important medicinal plants from the family Ranunculaceae, and its rhizome has been widely used in Traditional Chinese Medicine for centuries. Here, we analyzed the chemical components and the transcriptome of the Chinese goldthread from three biotopes, including Zhenping, Zunyi and Shizhu. We built comprehe...

Protective effects of alginate–chitosan microspheres loaded with alkaloids from Coptis chinensis Franch. and Evodia rutaecarpa (Juss.) Benth. (Zuojin Pill) against ethanol-induced acute gastric mucosal injury in rats

Zuojin Pill (ZJP), a traditional Chinese medicine formula, consists of Coptis chinensis Franch. and Evodia rutaecarpa (Juss.) Benth. in a ratio of 6:1 (w/w) and was first recorded in "Danxi's experiential therapy" for treating gastrointestinal disorders in the 15th century. However, the poor solubility of alkaloids from ZJP restricted the protective effect in treating gastritis and gastric ulce...

Inhibition of the bacterial surface protein anchoring transpeptidase sortase by isoquinoline alkaloids.

The inhibitory activity of Coptis chinensis rhizome-derived material was evaluated against sortase, a bacterial surface protein anchoring transpeptidase, from Staphylococcus aureus ATCC 6538p and compared to that of four commercially available isoquinoline alkaloids. The biologically active constituent of C. chinensis extract was characterized as the isoquinoline alkaloid, berberine chloride, b...
