Dataset on the identification of differentially expressed genes by annealing control primer-based PCR in mitochondrial DNA-depleted myocytes

چکیده
Changes in the mitochondrial DNA (mtDNA) content are believed to initiate a stress signal that leads to alterations in nuclear gene expression. This article presents data on the identification of nuclear genes that are expressed differentially in response to changes in the mtDNA content in myocytes using annealing controlled primers (ACP)-based PCR technology. The data obtained from L6 GLUT4myc myocytes showed that a total of 19 ACPs produced differentially expressed PCR amplicons in the mtDNA-depleted myocytes. Among those, 13 amplicons were cloned, sequenced, and identified successfully based on the GenBank database. To validate the efficacy of ACP-based PCR analysis, three differentially expressed genes (DEG10, 22 and 26) were confirmed by PCR using the specific primers. The further analysis and detailed results of DEG22 and its functional significance can be found in "C1q tumor necrosis factor alpha-related protein isoform 5 is increased in mitochondrial DNA-depleted myocytes and activates AMP-activated protein kinase." [1].

Annealing control primer system for identification of differentially expressed genes on agarose gels.
We developed GeneFishing technology, an improved method for the identification of differentially expressed genes (DEGs) using our novel annealing control primer (ACP) system. Because of high annealing specificity during PCR using the ACP system, the application of the ACP to DEG discovery generates reproducible, authentic, and long (100 bp to 2 kb) PCR products that are detectable on agarose ge...
متن کاملTracing the Pig and Cattle Origin in Processed Food and Feed Products Targeting Mitochondrial 12S rRNA Gene
Background: Species identification in commercially processed food and feed products is one of the important issues. This study was conducted to develop a genetic method for the detection of pig and cattle species in processed food and feed products using newly designed species-specific primers targeting mitochondrial 12S rRNA gene fragments. Methods: Two sets of specific primers were designed ...
متن کاملIdentification of drought induced differentially expressed genes in barley leaves using the annealing control-primer-based GeneFishing technique
In the present study, we have used an annealing-control-primer (ACP)-based differential display reverse transcription-polymerase chain reaction (RT-PCR) method to identify drought-stress-induced differentially expressed genes (DEGs) in barley leaves. Using 120 ACPs, a total of 12 up-regulated and 3 down-regulated genes were identified and sequenced. Temporal expression patterns of the 15 upor d...
متن کاملIdentification of differentially expressed genes in murine embryos at the blastocyst stage using annealing control primer system.
The identification of embryo-specific genes would provide insights into early embryonic development. However, the current methods employed to identify the genes that are expressed at a specific developmental stage are labor intensive and suffer from high rates of false positives. Here we employed a new and accurate reverse transcription-polymerase chain reaction (RT-PCR) method that involves an...
متن کاملEvaluation of differentially expressed genes identified in keratoconus
PURPOSE To identify the differentially expressed genes (DEGs) in the human keratocytes in keratoconus. METHODS Total RNA extracted from cultured corneal stromal fibroblasts from normal and keratoconic corneas were used for the synthesis of cDNA. DEGs were screened by an annealing control primer(TM)-based PCR method using GeneFishing() DEG kits. The differentially expressed bands were sequence...
