چکیده

Introduction Calcitonin-gene-related peptide (CGRP) is a 37-amino acid neuropeptide which was first identified in 1982 as a product of the calcitonin gene derived by alternative splicing [ 11. This original peptide is now referred to as a-CGRP; a second gene, unrelated to calcitonin, produces P-CGRP [2]. These two forms are closely related in both rat and man (Figure l ) , and are widely distributed throughout most parts of the nervous system. CGRP shares about 50% identity with another 37-amino acid peptide, amylin. This was first isolated from pancreatic amyloid plaques

International Union of Pharmacology. XXXII. The mammalian calcitonin gene-related peptides, adrenomedullin, amylin, and calcitonin receptors.

The calcitonin family of peptides comprises calcitonin, amylin, two calcitonin gene-related peptides (CGRPs), and adrenomedullin. The first calcitonin receptor was cloned in 1991. Its pharmacology is complicated by the existence of several splice variants. The receptors for the other members the family are made up of subunits. The calcitonin-like receptor (CL receptor) requires a single transme...

Identification of N-terminal receptor activity-modifying protein residues important for calcitonin gene-related peptide, adrenomedullin, and amylin receptor function.

Calcitonin-family receptors comprise calcitonin receptor-like receptor (CL) or calcitonin receptor and receptor activity-modifying protein (RAMP) pairings. Calcitonin gene-related peptide (CGRP) receptors are CL/RAMP1, whereas adrenomedullin (AM) receptors are CL/RAMP2 (AM1 receptor) or CL/RAMP3 (AM2 receptor). Amylin (Amy) receptors are RAMP hetero-oligomers with the calcitonin receptor (AMY1,...

Heterodimers and family-B GPCRs: RAMPs, CGRP and adrenomedullin.

RAMPs (receptor activity-modifying proteins) are single-pass transmembrane proteins that associate with certain family-B GPCRs (G-protein-coupled receptors). Specifically for the CT (calcitonin) receptor-like receptor and the CT receptor, this results in profound changes in ligand binding and receptor pharmacology, allowing the generation of six distinct receptors with preferences for CGRP (CT ...

Effects of amylin and adrenomedullin on the skeleton.

Amylin and adrenomedullin are related peptides with some homology to both calcitonin and calcitonin gene-related peptide (CGRP). All these peptides have in common a 6-amino acid ring structure at the amino-terminus created by a disulfide bond. In addition, the carboxy-termini are amidated. Both amylin and adrenomedullin have recently been found to stimulate the proliferation of osteoblasts in v...

International Union of Pharmacology. LXIX. Status of the calcitonin gene-related peptide subtype 2 receptor.

Historically, calcitonin gene-related peptide (CGRP) receptors have been divided into two classes, CGRP(1) and CGRP(2). After the cloning of calcitonin receptor-like receptor (CLR) and receptor activity-modifying proteins (RAMPs), it became clear that the CGRP(1) receptor was a complex between CLR and RAMP1. It is now apparent that the CGRP(2) receptor phenotype is the result of CGRP acting at ...
