Classification of binding preferences for 1000 novel transcription factors synthesized using a wheat germ cell-free translation system and Biacore analysis

چکیده

Label-free interaction analysis using BiacoreTM systems, combined with a highly efficient cell-free translation system provides a powerful approach to establish the targets of novel transcription factors. Interactions between 1000 proteins and 53 DNA oligonucleotide sequences were performed directly from in vitro translation microplates, greatly simplifying DNA binding analysis compared to traditional gel-based methods. Biacore assays enabled transcription factors to be classified by their sequence preferences and revealed a wide range of selectivities.
