چکیده

PURPOSE: Mesenchymal Stromal Cells (MSCs) are a promising source for cell therapy due to their proregenerative and immunomodulatory effects. These properties can be enhanced by engineering MSCs to overexpress and secrete growth factors favorable to wound healing processes. Here, we aimed to develop a platform for targeted gene insertion in human bone marrow (hBM)-MSCs that utilizes CRISPR/Cas9 nuclease and recombinant adeno-associated virus serotype 6 (rAAV6) to deliver a designed homologous recombination-based repair template. Using this method, we engineered hBM-MSCs to overexpress and secrete soluble platelet derived growth factor B (PDGF-B) and examined the ability of this cell line to promote wound healing.

Simvastatin combined with bone marrow mesenchymal stromal cells (BMSCs) improve burn wound healing by ameliorating angiogenesis through SDF-1α/CXCR4 pathway

Objective(s): Chemokines are wound mediators that promote angiogenesis during wound healing. We hypothesized that Simvastatin in combination with the bone marrow mesenchymal stromal cells (BMSCs) improve burn wound healing by ameliorating angiogenesis via SDF-1α/CXCR4 pathway.Materials and Methods: Under general anesthesia, deep partial-...

Tumorigenic conversion of immortal human keratinocytes through stromal cell activation.

The stromal microenvironment plays a crucial role in tumor development and progression. One of the most potent activators of stromal cells is the platelet-derived growth factor (PDGF). To investigate the role of PDGF in epithelial tumor development we stably transfected immortal nontumorigenic human keratinocytes with the PDGF-B cDNA. Transfected HaCaT cells overexpressed PDGF-B but remained ne...

Monitoring wound healing of burn in rat model using human Wharton’s jelly mesenchymal stem cells containing cGFP integrated by lentiviral vectors

Objective(s): Human Wharton’s Jelly mesenchymal stem cells (hWMSCs) are undifferentiated cells commonly used in regenerative medicine. The aim of this study was to develop a reliable tool for tracking hWMSCs when utilized as therapeutics in burnt disorders and also to optimize the cell-based treatment procedure. Materials and Methods: The hWMSCs were first isolated from fresh umbilical cord Wha...
