چکیده
Purtx~se: We like to demonstrate glycation of S,-, R,and y-crystallins in the microsectioned bovine lens, by isoelectric focusing using three staining methods. Methods: Pour bovine lenses of 1.185+0.131 years were frozen-sectioned into equa'cor and 11 layers. Watersoluble crvstallins were seuarated bv isoelectric focusing an8 stained: 1. with ?oomassie brilliant Blue (CBB); 2. with the lectin (LEc) Concanavalin-A, followed by horse-radish peroxidase (RRP) and diaminobenzidine (DAB): 3. for ulvcourotein receotors (REc). HRP and DAB. fiesuiis: crys&iiiis are separated i&o:'&, (I~-, R.-, a,-, 0, and y-crystallins. 1n thp lectin staining only BL-, R,and y-crystallins were positive, whereas the a&and I&-crystallins were? negative. Contrary to the glycated y-crystallins in the lens nucleus, the 13,and S,-crystallins were only glycated in the anterior cortex and to a somewhat lower extent also in the posterior cortical regions. Receptor staining was about 3 times less sensitive than the LEC! staining, but results were comparable. The degree of glycation: G, = LEC + CBB for y,-crystallins is mean 2.40,. for B,-crystallins 0.77 and for B,-crystallins 0.28. Conclusions: The degree of glycation of y-crystallins in the nucleus is 11 timeIs higher than cortical I?.,crystallins and 3.5 times higher than the cortical 13,crystallins. Application of Carver's model of chaperone activity gives the right interpretation of this study, because only glycated crystallins may fit into the central cavity (0 55A).

Disentangling the influence of insulin dependent diabetes mellitus on refraction.
The relation between the duration of insulin dependent, or juvenile onset, diabetes mellitus and changes in the refractive components of the eye undergoes a close and valuable assessment in the paper presented by Løgstrup et al in this month’s issue of the BJO (p 343). While it is known that the duration of diabetes has a marked eVect upon lens thickness 2 and the development of diabetic retino...
متن کاملComparisons of refractive errors between twins and singletons in Chinese school-age samples.
Studies have reported that refractive errors are associated with premature births. As twins have higher prevalence of prematurity than singletons, it is important to assess similarity of the prevalence of refractive errors in twins and singletons for proper interpretations and generalizations of the findings from twin studies. We compared refractive errors and diopter hours between 561 pairs of...
متن کاملLens thickness and insulin dependent diabetes mellitus: a population based twin study.
AIM To investigate the relation between lens thickness and duration of insulin dependent diabetes mellitus (IDDM). METHODS From the new population based Danish twin register, containing 20,888 twin pairs born between 1953 and 1982 (inclusive), all twin pairs having one or both partners affected with IDDM were searched. Among the 45 twin pairs available for clinical eye examination there were ...
متن کاملFactors aVecting pupil size after dilatation: the Twin Eye Study
Background/aims—Well dilated pupils make eye surgery easier. A classic twin study was established to examine the relative importance of genes and environment in the variance of pupil size after mydriasis, and to examine the eVects of other factors such as age, iris colour, and refractive error. Methods—506 twin pairs, 226 monozygotic (MZ) and 280 dizygotic (DZ), aged 49–79 (mean age 62.2 years,...
