Acrylamide gel electrophoresis studies of the incorporation of cytidine- 3 H into mouse skin RNA at early times after treatment with phorbol esters.

چکیده
The nature of the early RNA response was analyzed by polyacrylamide gel electrophoresis. The extraction procedure allowed quantitative isolation of undegraded RNA from whole mouse skin. By 3 hr after treatment with effective tumor promoters, there was a large stimulation of incorporation of cytidine-3H into "32 S" RNA, rRNA, and 4 to 5 S RNA. Stimulation of cytidine-3 H incorporation into 32 S RNA was

Effects of the chemical irritants anthralin and benzoyl peroxide on mouse skin epithelial cell protein production
The widely used topical agents anthralin and benzoyl peroxide are potent skin irritants. In the mouse skin model these compounds produce hyperplasia nd are effective skin tumor promoters. In the present studies, we have examined the effects of anthralin and benzoyl peroxide on epidermal proteins and compared them with changes produced by the potent phorbol ester tumor promoter 12-O-tetradecanoy...
متن کاملStimulation of the synthesis of mouse epidermal histones by tumor-promoting agents.
Topical application of 17 nmoles of the potent tumor promoter, 12-O-tetradecanoylphorbol-13-acetate, resulted in a stimulation of the incorporation of [(3)H]lysine into epidermal histones. Maximum incorporation occurred 24 hr after treatment, concurrent with maximum DNA synthesis. The effects of phorbol and two phorbol esters on histone synthesis were related to their tumor-promoting activities...
متن کاملMultiple effects of dexamethasone on protein synthesis and hyperplasia caused by a tumor promoter.
Dexamethasone efficiently inhibits the promotion of tu mors in mouse skin by 12-0-tetradecanoyl-phorbol-13-acetate (TPA) and reduces TPA-induced hyperplasia, as determined by histolÃ3gica! examination and rate of incorporation of labeled precursors into macromolecules of mouse epidermis. When the protein complement of mouse epidermal cytosol was assayed by electrophoresis on acidic polyacrylami...
متن کاملNerve growth factor regulates both the phosphorylation and steady-state levels of microtubule-associated protein 1.2 (MAP1.2)
This study characterizes effects of nerve growth factor (NGF) on the steady-state level and phosphorylation of a high molecular mass microtubule-associated protein in PC12 rat pheochromocytoma cells. Past work showed that NGF significantly raises the relative levels of this phosphoprotein, designated MAP1.2, with a time course similar to that of neurite outgrowth. To study this in greater detai...
متن کاملNeurogenic component of phorbol ester-induced mouse skin inflammation.
Tumor-promoting phorbol esters are potent inflammatory agents for mouse skin, and the potential mechanistic role of inflammation in tumor promotion is under active investigation. We have shown previously that resiniferatoxin, a uniquely irritant phorbol-related diterpene, acts as a capsaicin analogue to induce and then to block neurogenic inflammation. We report here that pretreatment of CD-1 m...
