چکیده

BACKGROUND Keratinocytes are the major constituent of epithelial cells at skin and mucosal surfaces such as oral mucosa. In our previous study, it was indicated that oral keratinocytes play active roles in oral mucosal adaptive immune response and an IL-alpha sub-network was identified. In the past decades IL-1, a revolutionarily ancient cytokine, has attracted our attention for its important roles in innate and adaptive immune responses. In this study we aim to show the IL-alpha sub-network in oral keratinocytes and explore its relevance in oral lichen planus (OLP). METHODS We first tested whether IL-1alpha regulated its sub-network genes including CXCL1, CXCL10, and ICAM1 mRNA levels in time and concentration dependent manners by real time PCR. Then we investigated the expression of IL-1alpha and CXCL1 in OLP tissues by immunohistochemical staining. RESULTS IL-1alpha regulated its sub-network genes including CXCL1, CXCL10, and ICAM1 mRNA levels in time dependent but not in concentration dependent manner. Immunohistochemistry studies showed that IL-1alpha and CXCL1 were expressed in OLP tissues, which were only detected in tissue transudate and whole unstimulated saliva in previous studies. CONCLUSIONS IL-1alpha regulates CXCL1, CXCL10, and ICAM1 in network form in oral keratinocytes. A complete characterization of the IL-1alpha sub-network will shed light on the exploration of IL-1 as the therapeutic target in OLP and help to illuminate the multiple regulatory functions of keratinocytes in oral mucosa or even in other mucosa sites.

Differential Expression of CXCL1, CXCL9 CXCL10 and CXCL12 Chemokines in Alopecia Areata

Background: Alopecia Areata (AA) is a non-scarring, autoimmune disorder which causes hair loss. Inflammatory reactions are involved in hair loss of the scalp and/or body. The involvement of chemokine receptors in the pathogenesis of AA is well defined among which, CXCL1 acts on neutrophils and CXCL9, CXCL10 and CXCL12 and serve as T lymphocytes recruiters. Objective: To study the serum levels o...

Alarmin function of cathelicidin antimicrobial peptide LL37 through IL-36γ induction in human epidermal keratinocytes.

Several dermatoses, including psoriasis, atopic dermatitis, and rosacea, alter the expression of the innate immune effector human cathelicidin antimicrobial peptide (CAMP). To elucidate the roles of aberrant CAMP in dermatoses, we performed cDNA array analysis in CAMP-stimulated human epidermal keratinocytes, the primary cells responding to innate immune stimuli and a major source of CAMP LL37 ...

Differential regulation of chemokines by IL-17 in colonic epithelial cells.

The IL-23/IL-17 pathway plays an important role in chronic inflammatory diseases, including inflammatory bowel disease. In inflammatory bowel disease, intestinal epithelial cells are an important source of chemokines that recruit inflammatory cells. We examined the effect of IL-17 on chemokine expression of HT-29 colonic epithelial cells. IL-17 strongly repressed TNF-alpha-stimulated expression...

Evaluation of circulating concentrations of CXCL1 (Gro-α), CXCL10 (IP-10) and CXCL12 (SDF-1) in ALL patients prior and post bone marrow transplantation.

The immune system plays an important role in the development of leukemia. CXC chemokines, as the molecular members of this system, are involved in the immune responses. Therefore, this study was designed to examine and compare the levels of CXCL1 (Gro-α), CXCL10 (IP-10) and CXCL12 (SDF-1) in ALL patients prior to and post bone marrow transplantation (BMT). In this experimental study, samples we...

Pediatric reference ranges for proinflammatory and anti-inflammatory cytokines in cerebrospinal fluid and serum by multiplexed immunoassay.

To define cytokine concentrations and detectability in children with noninflammatory neurological disorders (NIND). The multiplex bead assay technology was used for simultaneous measurement of 34 soluble cytokines/chemokines in cerebrospinal fluid (CSF) from 73 NIND. Sera from 36 healthy children and 37 NIND also were analyzed. In CSF, CXCL10 had the highest concentration; CCL2, CXCL10, and int...
