Development of Polymerase Chain Reaction assays with host-specific internal controls for Chlamydophila abortus

چکیده
Chlamydophila abortus (C. abortus) is one of the most important infectious agents causing abortion in ruminants. The bacterium is obligately intracellular, cannot grow on agar, but it needs cell culture or embryonated eggs for growth. Therefore, culture-independent detection methods such as the polymerase chain reaction are increasingly important and needed. The aim of this study was to develop a polymerase chain reaction assay with an internal control for detection of C. abortus in clinical samples. Using newly-designed two primer sets specific for C. abortus, the polymerase chain reaction assay was first tested with positive and negative control DNA and its sensitivity and specificity were determined. A new polymerase chain reaction protocol was developed by combining the new primer pair sets with bovine (12SM-FW and 12SBT-REV2) and ruminant host-specific primer sets (12S-FW and 12S-REV). In conclusion, the developed polymerase chain reaction assays can potentially be used for direct detection of Chlamydophila abortus in bovine and ruminant samples.

Development of New Modified Simple Polymerase Chain Reaction and Real-time Polymerase Chain Reaction for the Identification of Iranian Brucella abortus Strains
Brucellosis is primarily a worldwide zoonotic disease caused by Brucella species. The genus Brucella contains highly infectious species that are classified as biological threat agents. In this regard, the identification of Brucella can be a time-consuming and labor-intensive process posing a real risk of laboratory-acquired infection to the laboratory staff. This stud...
متن کاملComparisons of Polymerase Chain Reaction and Isolation in Cell Culture and Embryonated Hen’s Eggs for the Detection of Chlamydophila abortus in Aborted Ovine/Caprine Placenta
Enzootic abortion in ewes (EAE) is routinely diagnosed by the detection of elementary bodies of Chlamydophila abortus in placental smears stained with modified Ziehl-Neelsen’s (MZN) stain. The C. abortus can also be isolated from infected material in McCoy cells or embryonated eggs. Currently, the polymerase chain reaction (PCR) is increasingly being used for the detection and differentiation o...

BACKGROUND Reproductive disorders associated with chlamydial infection have been reported worldwide in cattle and there are indications of potential venereal transmission. METHODS Semen samples from 21 dairy bulls and cauda epididymidis tissue samples from 43 beef bulls were analysed for chlamydial agent by real-time polymerase chain reaction (PCR) including an internal amplification control ...

Abortion is one of the most important economic issues in sheep flocks. Chlamydophila abortus is an agent of enzootic abortions in sheep. Mycoplasma agalactiae is the main etiological agent of contagious agalactia, which can cause abortion in sheep. The aim of this study was to investigate the prevalence of M. agalactiae and C. abortus among aborted ovine fetu...
متن کاملComparison of Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) and Agglutination Assays in Diagnosis of Brucellosis in Golestan Province, North of Iran
Introduction: Brucellosis is one of the most common zoonotic infections worldwide. The clinical symptoms of brucellosis are similar to a wide range of diseases; hence, reliable diagnostic and laboratory methods are required to identify the causative agent. Iran is an endemic region of brucellosis, and many patients are misdiagnosed due to the nature of the infection. In this study, we aimed to ...
