Validation of the Sperm Class Analyser CASA System for Sperm Count in a Diagnostic Semen Analysis Laboratory
نویسندگان
چکیده
Sperm counts have been linked to several fertility outcomes making it an essential parameter of semen analysis. It has become increasingly recognised that Computer Assisted Semen Analysis provides improved precision over manual methods but systems are seldom robustly validated for use. The objective of this work was to gather the evidence to validate or reject the Sperm Class Analyser as a tool for routine sperm counting in a busy laboratory setting. The criteria examined were comparison with the Improved Neubauer and Leja 20μm chambers, within and between field precision, linearity from a stock diluted in both semen and media, accuracy against internal and external quality material, an assessment of uneven flow effects and a ROC analysis to predict fertility in comparison with the Neubauer method. The technology is not a stand alone “black box” but rather a tool for well trained staff that allow rapid, high number sperm counting providing errors are identified and corrected. The system will produce rapid, accurate, linear, precise results with less analytical variance than manual methods that correlate well against the Improved Neubauer chamber. The system provides superior predictive potential for diagnosing fertility problems.

متن کاملP-158: Effect of Occupational Exposureson Male Fertility
Background: Occupational exposures do have a negative impact on the male reproductive system, but sometimes it is difficult to isolate a single insult. Workers can be exposed to a number of harmful physical, chemical and psychological factors in their working environment. During recent years, the various diseases and disorders caused by these stressors have drawn the attention of a number of re...

متن کاملThe future of computer-aided sperm analysis
Computer-aided sperm analysis (CASA) technology was developed in the late 1980s for analyzing sperm movement characteristics or kinematics and has been highly successful in enabling this field of research. CASA has also been used with great success for measuring semen characteristics such as sperm concentration and proportions of progressive motility in many animal species, including wide appli...
متن کاملP-70: Effect of Xylose on Epididymal Sperm Motility in Water Buffalo (Bubalus bubalis)
Background: In mammalian spermatozoa, motility is one of the most important parameters for evaluating the quality of the semen. From a functional point of view, motility is strongly related to the spermatozoa’s ability to manage its energy status, Thus, modulation of energy status could be translated into significant changes in motility patterns. The aim of the present study was to evaluate xyl...
