Ovariectomized Cholesterol-Fed Rabbits

چکیده
Cardiovascular disease is a major killer in postmenopausal women in the industrialized societies. To investigate the effect of progestogen and 17p-estradiol replacement therapy on atherogenesis, we studied 60 cholesterol-fed ovariectomized rabbits for 13 weeks. They were randomly assigned to four groups of 15 rabbits each and received oral treatment with norethisterone acetate, levonorgestrel, 17f3-estradiol, or placebo. The active treatment groups achieved serum hormone concentrations, which produced physiological effects on the uterus. No significant differences in serum total cholesterol or ultracentrifuged lipoproteins (very low density, intermediate density, low density, or high density lipoproteins) were found between the four groups during the experimental period. There were no significant differences between the progestogen and placebo groups in the aortic accumulation of cholesterol. The estradiol group had only accumulated about half the aortic cholesterol as compared with the placebo group and the progestogen

Estrogen monotherapy and combined estrogen-progestogen replacement therapy attenuate aortic accumulation of cholesterol in ovariectomized cholesterol-fed rabbits.
Cardiovascular disease is currently the leading cause of death among women in the United States. To investigate the effect of postmenopausal hormone therapy on atherogenesis, we studied 75 cholesterol-fed female rabbits for 19 wk. The rabbits were randomly assigned to five groups. Four groups underwent bilateral ovariectomy followed by treatment with either 17 beta-estradiol, 17 beta-estradiol ...

متن کاملGender Difference in Endothelial Permeability of Aorta in Rabbits Consuming Normal or High-Cholesterol Diets
The endothelial permeability of aorta (EPA) is influenced by atherosclerosis. Whether the physiological difference in EPA is a sex-dependent phenomenon remains to be determined. The objective of this study was to determine the EPA difference between male and female rabbits. An experimental model was designed to obtain 4 groups of animals. The ovariectomized rabbits were fed with normal (gro...
متن کاملThe Effect of Estrogen on Endothelial Permeability of Aorta and the Level of Serum Nitrite Concentration in Cholesterol‑Fed Ovariectomized Rabbit
Estrogen Replacement Therapy (ERT) in postmenopausal women may decrease the risk of Coronary Artery Diseases (CAD). We hypothesized that Nitric Oxide (NO) releasing due to ERT may be the essential factor by which endothelial permeability decreases. Four groups of ovariectomized rabbits were under investigation for five weeks. Groups 1 & 4 received high cholesterol diet and other two groups (2 &...
متن کاملNorethindrone acetate enhances the antiatherogenic effect of 17beta-estradiol: a secondary prevention study of aortic atherosclerosis in ovariectomized cholesterol-fed rabbits.
The influence of progestogens in combination with 17beta-estradiol (E2) on cardiovascular disease remains controversial. This study investigated the effect of norethindrone acetate (NETA) combined with E2 on aortic atherosclerosis. Eighty mature female rabbits were ovariectomized, then fed a cholesterol-rich diet (240 mg/d) for 14 weeks to induce aortic atherosclerosis. They were randomized to ...
