چکیده

The isolation and study of glycerol-utilizing mutants of Rhodopseudomonas capsulata indicated that the wild-type organism has genes capable of coding for the catabolism of glycerol but is unable to express them. Furthermore, the genetic lesion in the original glycerol-utilizing mutant, L1, occurred very close to these genes.

Genetic-physical mapping of a photosynthetic gene cluster from R. capsulata.

Transposon mutagenesis and complementation analysis of the photosynthesis genes in Rhodopseudomonas capsulata is presented utilizing Tn5.7 mutagenized R-primes. The R-prime pRPS404 contains many of the genes necessary for the differentiation of the photosynthetic apparatus. Utilizing homologous recombination, 30 independent copies of Tn5.7 were inserted into the R. capsulata chromosome with sub...

Isolation and characterization of enhanced fluorescence mutants of Rhodopseudomonas capsulata.

After enrichment by a tetracycline suicide under conditions nonpermissive for the growth of mutants defective in photosynthesis, colonies were screened for enhanced fluorescence in near-infrared light by using high-speed infrared photography. Twenty mutants were isolated, and the chromatophore membranes were analyzed by a new, rapid microprocedure that revealed many different phenotypes among t...

The gene crtI mediates the conversion of phytoene into colored carotenoids in Rhodopseudomonas capsulata.

Carotenoids are membrane pigments present in all photosynthetic organisms, providing essential photoprotective functions. The first carotenoid formed in the pathway is phytoene, a colorless compound which is then converted into colored carotenoids by a series of dehydrogenation reactions. In the photosynthetic bacterium Rhodopseudomonas capsulata mutations that affect carotenoid biosynthesis be...

Anaerobic regulation of nitrogen-fixation genes in Rhodopseudomonas capsulata.

A Rhodopseudomonas capsulata nifH::lacZ gene fusion was used to isolate constitutive mutants of R. capsulata, unable to repress nif gene transcription anaerobically with every fixed-nitrogen source tested. When these nifc strains were grown aerobically, nif gene transcription was repressed. These results indicate that the regulation of nif gene transcription by fixed nitrogen is different from ...

Isolation and characterization of a glycerol auxotroph of Rhodopseudomonas capsulata: effect of lipid synthesis on the synthesis of photosynthetic pigments.

A glycerol auxotroph was isolated from Rhodopseudomonas capsulata for use as a system for studying membrane synthesis and function. When the mutant was deprived of glycerol, net phospholipid synthesis ceased immediately and a small amount of free fatty acids accumulated. A turnover of lipid occurred in both deprived and supplemented cultures. Deoxyribonucleic acid and protein synthesis continue...
