Sperm transcriptome profiling for assessment of boar semen freezability
نویسنده
چکیده
In this project transcriptome analysis on highthroughput RNA-sequencing (RNA-Seq) data of boar spermatozoa will be utilized to identify novel genetic markers associated with semen freezability. Three boars each with poor and good semen freezability will be subjected to transcriptome studies on sperm RNA-seq data, and in combination with bioinformatics screening, will select candidate genes associated with low or high sperm cryo-tolerance. A validation scheme will be used to enhance the analysis of the differentially expressed (DE) genes, putative markers for single nucleotide polymorphisms (SNPs) and the protein expression levels of the respective mRNA transcripts on a large animal population. Identification of sperm-derived genetic markers, associated with sperm freezability, will improve the cryopreservation technology of boar semen.

Short communication Isolation of total ribonucleic acid from fresh and frozen-thawed boar semen and its relevance in transcriptome studies
The main objective of this study was to isolate high-quality total ribonucleic acid (RNA) from raw fresh semen and frozen-thawed boar semen, using a protocol comprising the conventional TRIzol assay and a membrane-based technique, the PureLink RNA mini kit. Bioanalyzer profile revealed that the sperm RNA size distributions comprised mainly intact RNA ranging from 1500 to 1800 bp, without any de...
متن کاملPlenary contribution to International Conference on Boar Semen Preservation 2011. Genetic selection for freezability and its controversy with selection for performance.
Little data are available in the literature regarding freezability of boar sperm or its relationship with other traits. Existing data suggest the trait would respond favourably to selection, and information is available from other species suggesting components that might have changed. Genetic parameters are estimated for boar sperm freezability including heritability and correlations with other...
متن کاملIdentification of amplified restriction fragment length polymorphism markers linked to genes controlling boar sperm viability following cryopreservation.
This study investigated two hypotheses: 1) that consistent between-boar variation in frozen semen quality exists and is genetically determined, and 2) molecular markers linked to genes controlling semen freezability can be identified using amplified restriction fragment length polymorphism (AFLP) technology. Five ejaculates were collected from each of 129 boars. Semen was diluted into a commerc...
متن کاملState-of-the-art of boar sperm preservation in liquid and frozen state
Pig breeding is mainly conducted through Artificial Insemination (AI) in Western and developing countries. Apart from requiring specific catheters and trained staff, preserving boar semen in proper conditions is needed to ensure high reproductive performances. Although, at present, boar sperm may be preserved in liquid (15-17oC) or frozen states, more than 95% of AIs are conducted using liquid ...
متن کاملDetrimental Effects of Non-Functional Spermatozoa on the Freezability of Functional Spermatozoa from Boar Ejaculate
In the present study, the impact of non-functional spermatozoa on the cryopreservation success of functional boar spermatozoa was evaluated. Fifteen sperm-rich ejaculate fractions collected from five fertile boars were frozen with different proportions of induced non-functional sperm (0--native semen sample-, 25, 50 and 75% non-functional spermatozoa). After thawing, the recovery of motile and ...
