چکیده

Ilyobacter delafieldii produced an extracellular poly-[3-hydroxybutyrate (PHB) depolymerase when grown on PHB; activity was not detected in cultures grown on 3-hydroxybutyrate, crotonate, pyruvate or lactate. PHB depolymerase activity was largely associated with the PHB granules (supplied as growth substrate), and only 16 % was detected free in the culture supernatant. Monomeric 3-hydroxybutyrate was detectable as a product of depolymerase activity. The monomer was fermented to acetate, butyrate and H 2. After activation by coenzyme A transfer from acetyl-CoA or butyryl-CoA, the resultant 3-hydroxybutyryl-CoA was oxidized to acetoacetyl-CoA (producing NADH), followed by thiolytic cleavage to yield acetyl-CoA which was further metabolized to acetyl-phosphate, then to acetate with concomitant ATP production. The reducing equivalents (NADH) could be disposed of by the evolution of H 2, or by a reductive pathway in which 3-hydroxybutyrylCoA was dehydrated to crotonyl-CoA and reduced to butyryl-CoA. In cocultures of I. delafieldii with Desulfovibrio vulgaris on PHB, the H 2 partial pressure was much lower than in the pure cultures, and sulfide was produced. Thus interspecies hydrogen transfer caused a shift to increased acetate and H 2 production at the expense of butyrate.

Microbial degradation of natural and of new synthetic polymers.

In landfills, deposited waste material is usually faced with strictly anoxic conditions. This means that the design of new biodegradable polymers must take into consideration that degradation should be possible especially in the absence of molecular oxygen. Poly-beta-hydroxybutyrate is depolymerized by the anaerobic fermenting bacterium Ilyobacter delafieldii through an extracellular hydrolase....

Ilyobacter insuetus sp. nov., a fermentative bacterium specialized in the degradation of hydroaromatic compounds.

The mesophilic, anaerobic bacterium strain VenChi2T was isolated with quinic acid (1,3,4,5-tetrahydroxy-cyclohexane-1-carboxylic acid) as the sole source of carbon and energy. Of more than 30 substrates tested, only quinic acid and shikimic acid (3,4,5-trihydroxy-1-cyclohexene-1-carboxylic acid) were utilized, yielding acetate, propionate, butyrate, H2 and CO2 as fermentation products. Sugars, ...

Engineering a bzd cassette for the anaerobic bioconversion of aromatic compounds

Microorganisms able to degrade aromatic contaminants constitute potential valuable biocatalysts to deal with a significant reusable carbon fraction suitable for eco-efficient valorization processes. Metabolic engineering of anaerobic pathways for degradation and recycling of aromatic compounds is an almost unexplored field. In this work, we present the construction of a functional bzd cassette ...

Complete genome sequence of Ilyobacter polytropus type strain (CuHbu1T)

Ilyobacter polytropus Stieb and Schink 1984 is the type species of the genus Ilyobacter, which belongs to the fusobacterial family Fusobacteriaceae. The species is of interest because its members are able to ferment quite a number of sugars and organic acids. I. polytropus has a broad versatility in using various fermentation pathways. Also, its members do not degrade poly-β-hydroxybutyrate but...

Anaerobic degradation of 3-hydroxybenzoate by a newly isolated nitrate-reducing bacterium.

A Gram-negative nitrate-reducing bacterium, strain Asl-3, was isolated from activated sludge with nitrate and 3-hydroxybenzoate as sole source of carbon and energy. The new isolate was facultatively anaerobic, catalase- and oxidase-positive and polarly monotrichously flagellated. In addition to nitrate, nitrite, N2O, and O2 served as electron acceptors. Growth with 3-hydroxybenzoate and nitrate...
