چکیده

Cellulolytic enzyme preparations active against various forms of cellulose have been reported by a number of investigators who have used a variety of different sources (8). Fungi which have been employed for the preparation of such extracts are Sporotrichum cards (15), Aspergillus oryxae (3,2), Merulius lachrymans (9), and Coniophora cerebella (7), while bacteria have included Bacterium protoxoides (5), Bacterium bosporum (5), Cellulobacillus myxogenes (14), Cellulobacillus mucosus (14), Plectridium cellulolyticum (lo), several species of Cytophaga (l), and various thermophilic cellulose-decomposing bacteria (11). In addition, active cellulolytic extracts have been prepared from the intestinal juice of the snail Helix pomatia (6), from green malt (12), and from the protozoon Eudiplodinium neglectum which was isolated from cattle rumen (4). Although the data presented by several of these investigators are certainly suggestive of true cellulolytic enzyme activity, nevertheless these workers in general have not conclusively established the cell-free nature of their preparations. This is of considerable importance in view of failure of other investigators to obtain cell-free cellulolytic enzymatic preparation from fungi. Questions were raised as to the possibility of bacterial or fungal contamination during the period (usually several days or more) of testing for cellulolytic activity. The present paper describes the preparation and some of the properties of a cell-free cellulolytic enzyme from Myrothecium verrucaria, which has been shown to be one of the most active of the cellulose-destroying fungi isolated from deteriorated fabrics (16).

Induction of cellulase in Trichoderma viride as influenced by carbon sources and metals.

Cellulase is an adaptive enzyme in most fungi (Reese and Levinson, 1952), although it is constitutive in cellulolytic bacteria (Hammerstrom et al., 1955). Many polysaccharases are adaptive in fungi, including: pentosanase (Simpson, 1954), polygalacturonase (Phaff, 1947), chitinase (Reynolds, 1954), dextranase (Hultin and Nordstr6m, 1949), and xylanase and mannanase (S0rensen, 1952). Since many ...

Purification, characterization and decolorization of bilirubin oxidase from Myrothecium verrucaria 3.2190.

Myrothecium verrucaria 3.2190 is a nonligninolytic fungus that produces bilirubin oxidase. Both M. verrucaria and the extracellular bilirubin oxidase were tested for their ability to decolorize indigo carmine. The biosorption and biodegradation of the dye were detected during the process of decolorization; more than 98% decolorization efficiency was achieved after 7 days at 26°C. Additionally, ...

Characterisation of a Novel White Laccase from the Deuteromycete Fungus Myrothecium verrucaria NF-05 and Its Decolourisation of Dyes

A novel 'white' laccase was purified from the deuteromycete fungus, Myrothecium verrucaria NF-05, which was a high laccase-producing strain (40.2 U·ml(-1) on the thirteenth day during fermentation). SDS-PAGE and native-PAGE revealed a single band with laccase activity corresponding to a molecular weight of approximately 66 kDa. The enzyme had three copper and one iron atoms per protein molecule...
