Mitochondrial permeability transition pore: an enigmatic gatekeeper
نویسنده
چکیده
Mitochondrial permeability transition pore (MPTP) is a transient structure formed in the inner mitochondrial membrane (IMM) upon oxidative challenge of the organelle. The transition is a Ca 2+ -dependent increase of permeability of IMM leading to loss of transmembrane potential (∆ψm), mitochondrial swelling and, finally, rupture of outer mitochondrial membrane leading to release of factors causing apoptosis. Formation of this pore is an indication of the onset of necrotic cell death. In spite of several years of research, the molecular identity of this structure remains a mystery. The current review introduces MPTP, summarizes the research on various proteins that are thought to constitute the pore, enumerates factors that trigger pore formation, and the various experimental approaches that aid in understanding pore behavior.

Methanol extract and fraction of Anchomanes difformis root tuber modulate liver mitochondrial membrane permeability transition pore opening in rats
Objective: Extracts of Anchomanes difformis (AD) are used in folkloric medicine to treat several diseases and infections. However, their roles in mitochondrial permeability transition pore opening are not known. Material and Methods: The viability of mitochondria isolated from Wistar rat liver used in this experiment, was assessed by monitoring their swel...
متن کاملCKMT1 regulates the mitochondrial permeability transition pore in a process that provides evidence for alternative forms of the complex.
The permeability transition pore (PT-pore) mediates cell death through the dissipation of the mitochondrial membrane potential (ΔΨm). Because the exact composition of the PT-pore is controversial, it is crucial to investigate the actual molecular constituents and regulators of this complex. We found that mitochondrial creatine kinase-1 (CKMT1) is a universal and functionally necessary gatekeepe...
متن کاملMitochondrial permeability transition pore and postconditioning.
Postconditioning has recently been described as a powerful cardioprotection that prevents lethal reperfusion injury. Growing evidence suggests that mitochondrial permeability transition may be a key event in postconditioning. This proposition arises from the complementary observations that: (1) conditions for the mitochondrial permeability transition pore (mPTP) opening are built up during earl...
متن کاملCommentary: The m-AAA Protease Associated with Neurodegeneration Limits MCU Activity in Mitochondria
Mutations in subunits of mitochondrial m-AAA proteases in the inner membrane cause neurodegeneration in spinocerebellar ataxia (SCA28) and hereditary spastic paraplegia (HSP7). m-AAA proteases preserve mitochondrial proteostasis, mitochondrial morphology, and efficient OXPHOS activity, but the cause for neuronal loss in disease is unknown. We have determined the neuronal interactome of m-AAA pr...
متن کاملModulation of mitochondrial ion transport by inorganic polyphosphate - essential role in mitochondrial permeability transition pore
Inorganic polyphosphate (polyP) is a biopolymer of phosphoanhydride-linked orthophosphate residues. PolyP is involved in multiple cellular processes including mitochondrial metabolism and cell death. We used artificial membranes and isolated mitochondria to investigate the role of the polyP in mitochondrial ion transport and in activation of PTP. Here, we found that polyP can modify ion permeab...
