Synthesis of proteochondroitin sulfate by normal, nanomelic, and 5-bromodeoxyuridine-treated chondrocytes in cell culture.

چکیده
Chondrocytes grown in cell culture from micromelic chick sterna and 5-bromodeoxyuridine-treated normal cells showed reduced incorporation of sulfate into hyaluronidase-sensitive material as compared to normal chondrocytes. Proteochondroitin sulfate synthesized by normal embryonic chick chondrocytes in cell culture was heterogeneous on chromatography. The major chromatographic component was specifically inhibited in normal chondrocytes treated with 5-bromodeoxyuridine, and this component was absent in nanomelic chondrocytes.

Nanomelic chondrocytes synthesize a glycoprotein related to chondroitin sulfate proteoglycan core protein.
Chicken embryos homozygous for the autosomal recessive gene nanomelia exhibit cartilage defects, synthesize low levels of cartilage chondroitin sulfate proteoglycan (CSPG), and are missing the CSPG core protein (Argraves, W. S., McKeown-Longo, P. J., and Goetinck, P. F. (1981) FEBS Lett. 131, 265). In our studies of nanomelic chondrocytes in culture, we detected neither sulfate-labeled CSPG nor...
متن کاملCoordinate regulation of collagen and alkaline phosphatase levels in chick embryo chondrocytes.
Chick embryo tibial chondrocytes release into their extracellular matrix several species of proteochondroitin sulfate and collagen as well as matrix vesicles that are rich in Ca2+ and alkaline phosphatase and that appear to play a role in the calcification of cartilage. To determine whether there was any parallel regulation of the production of these products, the rates of collagen synthesis by...
متن کاملTransforming Growth Factor-β1 Preserves Bovine Nasal Cartilage against Degradation Induced by Interleukin-1α in Explant Culture
Background and Aims: Chondrocytes and their differentiation play a central role in joint diseases. Effect of the transforming growth factor (TGF)-β1 on chondrocyte characteristics and differentiation is not clearly understood. This study was undertaken to investigate the effects of TGF-β1 on tissue characteristics and morphology of chondrocytes against degradation induced by interleuk...
متن کاملQuantitative Analysis of the Proliferation and Differentiation of Rat Articular Chondrocytes in Alginate 3D Culture
Background: While articular chondrocytes are among those appropriate candidates for cartilage regeneration, the cell dedifferentiation during monolayer culture has limited their application. Several investigations have indicated the usefulness of alginate, but the topic of proliferation and differentiation of chondrocytes in alginate culture has still remained controversial. Methods: Rat articu...
متن کاملCostal Versus Articular Chondrocytes in Alginate Three-Dimensional Cultures
Given the difficulties in accessing articular cartilage as a source of chondrocytes to be used in fabricating cartilage constructs, alternative sources are required. The present study examined chondrocytes from costal cartilage for their suitability in cartilage tissue engineering. Chondrocytes isolated from rat knee and rib hyaline cartilage were separately mixed with alginate and placed in a ...
