The succinic dehydrogenase from Rhodospirillum rubrum.

چکیده
Succinic dehydrogenase has usually been associated with a macromolecular intracellular unit (mitochondria) whenever the enzyme has been isolated. In an ultracentrifugal analysis of the composition of extracts of various organisms, Schachman et al. (1952) found that Rhodospirillum rubrum yielded particles with a wide range of sedimentation constants. Succinic dehydrogenase has been demonstrated in extracts of R. rubrum by Vernon and Kamen (1953) using the reduction of cytochrome c or a dye as the assay, and by Eisenberg (1953) with a manometric assay. This article describes the properties of a partially purified succinic dehydrogenase system from R. rubrum that is associated with particles that have the same sedimentation constants as do the particles bearing pigments. A soluble bacterial succinic dehydrogenase has been obtained from Azotobacter vinelandii (Repaske, 1954).

Identification of two distinct lactate dehydrogenases in Rhodospirillum rubrum.
The activities of pyridine nucleotide-independent d- and l-lactate dehydrogenases were detected in membranes from Rhodospirillum rubrum grown under aerobic and phototrophic conditions. Crossed immunoelectrophoretic analysis revealed two antigenically distinct enzymes that were further distinguished by specificity for d- and l-stereoisomers of lactate and by the sensitivity of the d-lactate dehy...
متن کاملSuccinate dehydrogenase in Rhodopseudomonas sphaeroides: subunit composition and immunocross-reactivity with other related bacteria.
Antibodies were raised against the succinate dehydrogenase (SDH) present in the chromatophores of phototrophically grown Rhodopseudomonas sphaeroides. Crossed immunoelectrophoresis experiments indicated that the SDH present in the cytoplasmic membranes of heterotrophically grown R. sphaeroides is probably the same enzyme observed in the chromatophores. The enzyme was extracted by Triton X-100 i...
متن کاملSpectroelectrochemical characterization of the metal centers in carbon monoxide dehydrogenase (CODH) and nickel-deficient CODH from Rhodospirillum rubrum.
Carbon-monoxide dehydrogenase (CODH) from Rhodospirillum rubrum contains two metal centers: a Ni-X-[Fe4S4]2+/1+ cluster (C-center) that serves as the COoxidation site and a standard [Fe4S4]2+/1+ cluster (B-center) that mediates electron flow from the C-center to external electron acceptors. Four states of the C-center were previously identified in electron paramagnetic resonance (EPR) and Mössb...
متن کاملHomoserine Dehydrogenase of Rhodospirillum rubrum
Homoserine dehydrogenase catalyzes the reductive conversion of aspartate P-semialdehyde to homoserine (l), which has been identified in several microorganisms as a precursor of three other amino acids. On the one hand, homoserine is converted to methionine, and through a separate sequence of reactions is transformed to threonine, a precursor of isoleucine (2). Recent studies (3-7) have provided...
متن کاملCarbon monoxide dehydrogenase from Rhodospirillum rubrum.
The carbon monoxide dehydrogenase from the photosynthetic bacterium Rhodospirillum rubrum was purified over 600-fold by DEAE-cellulose chromatography, heat treatment, hydroxylapatite chromatography, and preparative scale gel electrophoresis. In vitro, this enzyme catalyzed a two-electron oxidation of CO to form CO2 as the product. The reaction was dependent on the addition of an electron accept...
