BIOPHARMACEUTICS DRUG DISPOSITION CLASSIFICATION SYSTEM: AN EXTENSION OF BIOPHARMACEUTICS CLASSIFICATION SYSTEM Kataria

چکیده
The Biopharmaceutics Classification System (BCS) was developed for prediction of in vivo pharmacokinetic performance of drug products from measurements of permeability and solubility. If the permeability criteria changed with metabolism criteria than it may be useful in predicting overall drug disposition, including routes of drug elimination and the effects of efflux and absorptive transporters on oral drug absorption, importance of food effects, and transporter effects on post absorption systemic drug concentrations following oral and intravenous dosing known as Biopharmaceutics drug disposition classification system (BDDCS). The drugs that have high permeability but poor metabolism are generally hydrophilic molecules with low molecular weight are likely to be absorbed by active transport mechanisms. Suggesting that drugs have extensive metabolism are highly absorbed. BDDCS using elimination criteria may expand the number of Class 1 drugs eligible for a biowaiver of in vivo bioequivalence (BE) studies and provide predictability of drug disposition profiles for Classes 2, 3, and 4 compounds.

Distinguishing between the Permeability Relationships with Absorption and Metabolism To Improve BCS and BDDCS Predictions in Early Drug Discovery
The biopharmaceutics classification system (BCS) and biopharmaceutics drug distribution classification system (BDDCS) are complementary classification systems that can improve, simplify, and accelerate drug discovery, development, and regulatory processes. Drug permeability has been widely accepted as a screening tool for determining intestinal absorption via the BCS during the drug development...
متن کاملBioequivalence of oral products and the biopharmaceutics classification system: science, regulation, and public policy.
The demonstration of bioequivalence (BE) is an essential requirement for ensuring that patients receive a product that performs as indicated by the label. The BE standard for a particular product is set by its innovator, and this standard must subsequently be matched by generic drug products. The Biopharmaceutics Classification System (BCS) sets a scientific basis for an improved BE standard fo...

Identifying molecules that interact with P-glycoprotein (P-gp) is important for drug discovery but is also generally reliant on time-consuming in vitro and in vivo studies. As an alternative approach, the current study applied pharmacophore models and database screening to rapidly retrieve molecules that bind as substrates or inhibitors for P-gp from commercial databases and then confirmed thei...
متن کاملImatinib metabolism and disposition in isolated rat perfused liver
Imatinib is an orally administered tyrosine kinase inhibitor which inhibits the Bcr-Abl protein-tyrosine kinase with high selectivity. Imatinib is rapidly absorbed from the gut, after oral intake and has an almost absolute bioavailability of 98%. The metabolism of imatinib is mediated by the cytochrome P450 (CYP) isoenzymes in the liver and gut wall. CGP74588 is a major active metabolite of ima...

Specific biopharmaceutics classification investigation and study on phamacokinetic profile of a novel drug candidate (2-methylcarbamoyl-4-{4-[3- (trifluoromethyl) benzamido] phenoxy} pyridinium 4-methylbenzenesulfonate monohydrate, NCE) were carried out. Equilibrium solubility and intrinsic dissolution rate (IDR) of NCE were estimated in different phosphate buffers. Effective intestinal permeab...
