چکیده

Active germinal centers are recognized as areas of rapidly proliferating blast cells in the white pulp of the spleen and in the cortex of lymph nodes. The origin and life history of these cells, their relationship to the surrounding mantle zone of small lymphocytes and, in particular, the functional significance of these two groups of cells is still unknown. Flemming (1) has proposed that germinal centers are a major site of production of small lymphocytes, but this interpretation has been disputed by others (2, 3). Hellman (4) has suggested that these structures are a reaction site against invading organisms. This is consistent with the fact that germinal centers appear specifically after exposure to antigen (5, 6). Rapid proliferation of blast cells in the germinal centers develops later than the peak of the primary response and coincides with development of primed cells after exposure to antigen (7, 8). Moreover, it can be shown by microdissection methods that the cells capable of giving a secondary response develop in the splenic white pulp at this time (9, 10). I t is therefore important to determine the life history and immunological properties of the cell types arising in the white pulp after intravenous injection of antigen.

Relationship of Germinal Centers in Lymphoid Tissue to Immunological Memory

The fate, proliferation, and developmental potentialities of cell suspensions made from white pulp containing large germinal centers have been studied in the mouse by transfer of cells labeled with thymidine-(3)H to lethally irradiated, syngeneic recipients. Radioautographic analyses were made using both smears and sections of a variety of tissues. Thymidine-(3)H-labeling patterns of white pulp...

Extrafollicular antibody responses.

In adaptive antibody responses, B cells are induced to grow either in follicles where they form germinal centers or in extrafollicular foci as plasmablasts. Extrafollicular growth typically occurs in the medullary cords of lymph nodes and in foci in the red pulp of the spleen. It is not a feature of secondary lymphoid tissue associated with the internal epithelia of the body. All types of naïve...

ارزیابی تغییرات سلولی بافت طحال و تغییرات سرولوژیک خون محیطی در اتوترانسپلنتیشن طحال در مقایسه با موارد اسپلنکتومی شده در رت

Background & Aims: Spleen is one of the most common sites of injury after trauma. When splenectomy due to spleen damage is unavoidable, autotransplantation of splenic tissue is the only possible option to preserve splenic function. Immunologic function of spleen against infections is unremarkable but compensation levels of immunological activity and structural rearrangement of transplanted spl...

HIV-specific effector cytotoxic T lymphocytes and HIV-producing cells colocalize in white pulps and germinal centers from infected patients.

Human immunodeficiency virus (HIV) infection is characterized by the massive infiltration of secondary lymphoid organs with activated CD8(+) T lymphocytes. While converging data indicated that these cells were HIV-specific cytotoxic T lymphocytes (CTLs) responsible for HIV spread limitation, direct evidence was lacking. Here, the presence of HIV-specific effector CTLs was demonstrated directly ...

T Helper Cells in Murine Germinal Centers Are Antigen - specific Emigrants That Downregulate Thy -

After immunization, activated splenic T cells proliferate in periarteriolar lymphoid sheaths (PALS) and subsequently migrate to the lymphoid follicle where they enter nascent germinal centers. Analysis o f T C R V(D)J gene rearrangements indicates extensive emigration, frequently involving more than a single white pulp region. These migrants constitute a unique set of T helper ceils that expres...
