Inhibition of 2,4-dichlorophenoxyacetic Acid-stimulated elongation of pea stem segments by a xyloglucan oligosaccharide.

چکیده
Xyloglucan, isolated from the soluble extracellular polysaccharides of suspension-cultured sycamore (Acer pseudoplatanus) cells, was digested with an endo-beta-1,4-glucanase purified from the culture fluid of Trichoderma viride. A nonasaccharide-rich Bio-Gel P-2 fraction of this digest inhibited 2,4-dichlorophenoxyacetic-acid-stimulated elongation of etiolated pea stem segments. The inhibitory activity of this oligosaccharide fraction exhibited a well-defined concentration optimum between 10(-2) and 10(-1) micrograms per milliliter. Another fraction of the same xyloglucan digest, rich in a structurally related heptasaccharide, did not, at similar concentrations, significantly inhibit the elongation.

Suppression and acceleration of cell elongation by integration of xyloglucans in pea stem segments.
Xyloglucan is a key polymer in the walls of growing plant cells. Using split pea stem segments and stem segments from which the epidermis had been peeled off, we demonstrate that the integration of xyloglucan mediated by the action of wall-bound xyloglucan endotransglycosylase suppressed cell elongation, whereas that of its fragment oligosaccharide accelerated it. Whole xyloglucan was incorpora...
متن کاملOccurrence of cello-oligosaccharides in the apoplast of auxin-treated pea stems
Treatment of pea (Pisum sativum L.) hypocotyl segments with indole-3-butyric acid, which promotes segment elongation, increased the solubilization of both xyloglucan and cello-oligosaccharides in the apoplast of auxin-treated pea stems. The cello-oligosaccharides were isolated from the apoplastic solution with a charcoal/Celite column and were identified as cellobiose, cellotriose, and cellotet...
متن کاملAuxin and the response of pea roots to auxin transport inhibitors: morphactin.
The auxin transport inhibitor methyl-2-chloro-9-hydroxyfluorene-9-carboxylate (CFM), a morphactin, inhibits negative geotropism, causes cellular swelling, and induces root hair formation in roots of intact Pisum sativum L. seedlings. In excised pea root tips, CFM inhibits elongation more than increase in fresh weight (swell ratio = 1.3 at 20 mum CFM). CFM growth inhibition was expressed in the ...
متن کاملRelationship between Promotion of Xyloglucan Metabolism and Induction of Elongation by Indoleacetic Acid.
Auxin promotes the liberation of a xlyoglucan polymer from the cell walls of elongating pea (Pisum sativum) stem segments. The released polymer can be isolated from the polysaccharide fraction of the water-soluble portion of tissue homogenates, thus providing as assay for this kind of metabolism. Promotion of xyloglucan metabolism by auxin begins within 15 minutes of hormone presentation. The e...
متن کاملInfluence of 2,4-Dichlorophenoxyacetic Acid on Pathways of Glucose Utilization in Bean Stem Tissues.
2,4-Dichlorophenoxyacetic acid (2,4-D) is the best known selective herbicide for effective control of broadleafed weeds in monocotyledonous crops. Although the mechanisms by which 2,4-D acts on plants have not been elucidated, several lines of investigation have shown: A. Stimulatory and inhibitory effects on the respiration of pea and oat tissues, depending upon the concentrations employed (6)...
