چکیده

PEGylated peptides and proteins generate signifi cant challenges for the detailed structural characterization of biotherapeutics, mainly due to polyethylene glycol (PEG) heterogeneity. This application note describes a method for analyzing PEG and PEGylated protein using an Agilent HPLC coupled to an Agilent 6520 Accurate Mass Q-TOF LC/MS System. A charge stripping agent (triethylamine) was used as a post column addition to aid in obtaining a simpler mass spectrum and hence a better interpretation of the results. The results showed signifi cant improvement in the mass spectrum quality for PEG and the PEGylated protein. Authors Ravindra Gudihal and Suresh Babu CV Agilent Technologies India Pvt. Ltd. Reproducible preparation and effective separation of PEGylated recombinant human granulocyte colony-stimulating factor with novel "PEG-pellet" PEGylation mode and ion-exchange chromatography.

A novel preparation for polyethylene glycol (PEG) derivatives and chromatographic separation procedure of the PEGylated recombinant human granulocyte colony-stimulating factor (rhG-CSF) were designed to evaluate the reproducibility and scalability at large laboratory-scale level. The new "PEG-pellet" PEGylation mode was successfully applied to control the pH fluctuation during the conjugation r...

Pharmacokinetic disposition of polyethylene glycol-modified salmon calcitonins in rats.

This study first reports the pharmacokinetic disposition of polyethylene glycol (PEG)-modified salmon calcitonin (sCT) based on the number of attached PEG molecules. PEG-modified sCT was prepared by covalent linkage with succinimidyl carbonate monomethoxy polyethylene glycol. Mono- and di-PEG-sCTs were separated by size exclusion and reverse phase HPLC, and radioiodinated by the chloramine-T me...

A Better Anti-Diabetic Recombinant Human Fibroblast Growth Factor 21 (rhFGF21) Modified with Polyethylene Glycol

As one of fibroblast growth factor (FGF) family members, FGF21 has been extensively investigated for its potential as a drug candidate to combat metabolic diseases. In the present study, recombinant human FGF21 (rhFGF21) was modified with polyethylene glycol (PEGylation) in order to increase its in vivo biostabilities and therapeutic potency. At N-terminal residue rhFGF21 was site-selectively P...

This Application Note used hen egg white lysozyme as a model protein for purifi cation and biophysical characterization of PEGylated proteins. PEGylation to the N-terminus of lysozyme in a site-specifi c manner was achieved using PEG-propionaldehyde derivatives with a molecular weight of Mw 20,000. A semi-preparative RP-HPLC method for the purifi cation of PEGylated lysozyme was developed using...

Using anti-poly(ethylene glycol) bioparticles for the quantitation of PEGylated nanoparticles

Attachment of polyethylene glycol (PEG) molecules to nanoparticles (PEGylation) is a widely-used method to improve the stability, biocompatibility and half-life of nanomedicines. However, the evaluation of the PEGylated nanomedicine pharmacokinetics (PK) requires the decomposition of particles and purification of lead compounds before analysis by high performance liquid chromatography (HPLC), m...
