Conjoined NO-sensitive guanylyl cyclases

چکیده
Background NO-sensitive guanylyl cyclases (NO-GC’s) that catalyze the reaction of GTP to the second messenger molecule cGMP are heterodimeric enzymes consisting of an a and a b1 subunit. The two prevalent isoforms in humans (a1/b1, a2/b1) are time-tested targets for drugs that release nitric oxide (NO) and new compounds that either sensitize the enzyme for activation by NO or activate the enzyme independently of NO. Homologous NO-GC subunits consist of an amino-terminal HNOX domain followed by a PASand a coiled coil domain and the carboxy-terminal catalytic domain.

Particulate guanylyl cyclases: multiple mechanisms of activation.
Cyclic GMP (cGMP), a key messenger in several signal transduction pathways, is synthesized from GTP by a family of enzymes termed guanylyl cyclases, which are found in two forms: cytosolic (soluble) and membrane-bound (particulate). The past decade has brought significant progress in understanding the molecular mechanisms that underlie the regulation of particulate guanylyl cyclases and new mem...
متن کاملGuanylyl cyclases across the tree of life.
This review explores the origins, diversity and functions of guanylyl cyclases in cellular organisms. In eukaryotes both cGMP and cAMP are produced by the conserved class III cyclase domains, while prokaryotes use five more unrelated catalysts for cyclic nucleotide synthesis. The class III domain is found embedded in proteins with a large variety of membrane topologies and other functional doma...
متن کاملMsGC-beta3 forms active homodimers and inactive heterodimers with NO-sensitive soluble guanylyl cyclase subunits.
Soluble guanylyl cyclases are typically obligate heterodimers, composed of a single alpha and a single beta subunit. MsGC-beta3, identified in the tobacco hornworm Manduca sexta, was the first example of a soluble guanylyl cyclase that exhibited enzyme activity without the need for coexpression with additional subunits. Subsequent studies have revealed that the mammalian beta2 subunit also shar...
متن کاملGuanylyl cyclases and signaling by cyclic GMP.
Guanylyl cyclases are a family of enzymes that catalyze the conversion of GTP to cGMP. The family comprises both membrane-bound and soluble isoforms that are expressed in nearly all cell types. They are regulated by diverse extracellular agonists that include peptide hormones, bacterial toxins, and free radicals, as well as intracellular molecules, such as calcium and adenine nucleotides. Stimu...
متن کاملNO signalling in synaptic transmission
Background The NO/cGMP signaling cascade has been proposed to play a role in long-term potentiation (LTP) and the modulation of synaptic transmission. Nitric oxide is formed enzymatically by NO synthases (NOS); two NOS, the endothelial and neuronal isoform (eNOS, nNOS) produce NO as a signalling molecule. Functionally, NO has been reported to act as a retrograde messenger that is generated post...
