Isolation of Glutamic Acid Methyl Ester from anEscherichia coZi Membrane Protein Involved in Chemotaxis*
نویسندگان
چکیده
We have isolated glutamic acid 5-methyl ester from an Escherichia coli protein that is involved in chemotaxis. The bacteria were first incubated with [methyl-“Hlmethionine under conditions which are known to result in methylation of the protein. The protein, isolated by gel electrophoresis, was then digested by successive treatment with three proteolytic enzymes. One of the products was [methyl-“Hlglutamic acid 5-methyl ester, identified by comparison with an authentic sample in the following studies: (a) chromatography on an automatic amino acid analyzer, (6) chromatography on paper in two solvent systems, (c) chromatography on paper of the N-acetyl derivatives, and (d) stability of the ester bond to various pH conditions. No aspartic acid 4methyl ester was found in the enzymatic digest. Treatment of the methylated protein with alkali released the radioactivity as [“HImethanol, which was identified by gas chromatography and by preparation of the 3,5-dinitrobenzoate.

Background: Lipase is an enzyme with immense application potential. Ester synthesis by lipase catalysis in organic media is an area of key industrial relevance. Enzymatic preparations with traits that cater to the needs of this function are hence being intensely researched. Objective: The objectives of the study were to immobilize the lipase from Bacillus sp. PS35 by cross-linking and adsorpti...
متن کاملMethylation-independent aerotaxis mediated by the Escherichia coli Aer protein.
Aer is a membrane-associated protein that mediates aerotactic responses in Escherichia coli. Its C-terminal half closely resembles the signaling domains of methyl-accepting chemotaxis proteins (MCPs), which undergo reversible methylation at specific glutamic acid residues to adapt their signaling outputs to homogeneous chemical environments. MCP-mediated behaviors are dependent on two specific ...
متن کاملPosttranslational processing of methyl-accepting chemotaxis proteins in Escherichia coli.
Methyl-accepting chemotaxis proteins (MCPs) of Escherichia coli undergo changes in methylation state in response to chemical stimuli. The addition of methyl groups to MCP is dependent on cheR function; their removal is dependent on cheB function. This MCP methylation system is instrumental in establishing the unstimulated swimming pattern of E. coli and in enabling the cell to carry out sensory...
