Determination of organic iodine compounds in serum. IV. A new nonincineration technic for serum thyroxine.

چکیده
Unidentified substances in acetic acid column eluates, obtained in the determination of serum thyroxine by column chromatography with the anion-exchange resin Dowex-1, prevented the direct determination of thyroxine by interference with the iodidecatalyzed ceric-arsenite reaction. Pretreatment of the eluates with Br2 or Cl2 eliminated the interference and permitted the direct assay of thyroxine by the cericarsenite system without incineration or wet digestion. Pretreatment with Br2 or Cl2 also significantly enhanced the catalytic activity of thyroxine, rendering its iodine nearly equivalent to inorganic iodide in the cericarsenite system. This nonincineration thyroxine assay is significantly more specific than methods that employ wet or dry ashing, because many organic iodine compounds used in diagnosis and therapy are not measured. A comparison of the nonincineration technic with the alkaline dry-ash method on 110 random serums revealed a mean difference of 0.13 .g./100 ml. (X = 4.5 g./ 100 ml.), with the nonincineration technic yielding the higher result. This difference was found to be statistically significant (p < 0.001).

Protein-bound iodine determination by activation analysis.
The most commonly-used test for determining the serum concentration of thyroid hormones is the protein-bound iodine analysis (PBI) which measures the serum's entire organic and some inorganic iodide concentration. Since it is desired to measure only that portion of the serum iodine which is in the thyroxine molecule, this test may be misleading. The method of PBI determination rou tinely used i...

SEVERAL TESTS have been proposed for determining the gross contamination sometimes encountered in serums submitted for protein-bound iodine analysis (1-5). These screens are all l)ased on the use of the coupled Ce(IV-As(III)) reaction as a detection device. However, ceric ions in strong acid solution represent a powerful oxidation system for many organic compounds (6). There is, therefore, a go...
متن کاملConcentration of serum total iodine and thyroid hormones in Holstein cows in central Iran
The aim of this study was to evaluate total iodine, triiodothyronine (T3) and thyroxine (T4) status in the blood serum of Holstein cows on six farms in central Iran. Total iodine, T3 and T4 concentrations were measured in 90 blood serum samples (15 per farm). Mean concentrations of total iodine (38.80 ± 1.52 µg/l) and T3 (1.57 ± 0.10 nmol/l) were lower but the mean concentration of T4 (45.60 ± ...
متن کاملSimplified technic for the determination of serum protein-bound iodine.
A simplified technic for the determination of serum protein-bound iodine (PBI) is presented. Endogenouslylabeled 1131 PBI is completely recovered. The technic is basedon the method of Barker et al. (1) and includes the following modifications: The number of washesof the precipitated serum proteins are decreased. Unashed iodide standards are used in a sodium carbonate solution rather than an int...
متن کاملDetermination of organic iodine compounds in serum. II. Chromatographic studies of exogenous organic iodine compounds affecting the PBI and BEI.
R1_ values of organic iodine compounds which interfere with the determination of serum protein-bound iodine (P81) and butanol-extractable iodine (BEI) levels were obtained for the solvent system 2-butanol-4% NH4OH (3:1) utilizing descending paper chromatography. The feasibility of employing this system for identification of exogenous organic iodine in patients’ sera was investigated. In general...
