چکیده

L.N. MANCHESTER, A . TOOLE AND R . GOODACRE. 1995. Forty-eight strains of Carnobacter ium were examined by pyrolysis mass spectrometry (PyMS). T h e effects of culture age and reproducibility over a 4 week period were also examined. T h e results were analysed by multivariate statistical techniques and compared with those from a previous numerical taxonomic study based on morphological, physiological and biochemical characteristics and with studies which used DNA-DNA and 16s rRNA sequence homologies. Taxonomic correlations were observed between the PyMS data and the previous studies. Culture age was observed to have little effect on the mass spectra obtained and the reproducibility study indicated that there was very little variation over the 4 week period. I t was concluded that PyMS provides a reliable method for studying carnobacterial classification and provides a rapid way for clarifying and refining subgeneric relationships within the genus Carnobacter ium. Further work may also show that it offers a potentially very rapid and accurate method for the identification of Carnobacter ium.

Whole-organism fingerprinting of the genus Carnobacterium using Fourier transform infrared spectroscopy (FT-IR).

Sixty seven strains of Carnobacterium, atypical Lactobacillus, Enterococcus durans, Lactobacillus maltaromicus and Vagacoccus salmoninarum were examined by Fourier transform infrared (FT-IR) spectroscopy. The effects of culture age and reproducibility over a six month period were also investigated. The results were analysed by multivariate statistics and compared with those from a previous nume...

Chemical Characterization of Recent and Buried Woods by Analytical Pyrolysis Comparison of Pyrolysis Data with C Nmr and Wet Chemical Data

In a previous study, selective degradation of carbohydrates versus lignin in buried woods was demonstrated by non-destructive spectroscopy (C NMR) and destructive wet chemical methods (acid hydrolysis and alkaline CuO oxidation). In this paper, spruce, alder and oak woods deposited in coastal sediments and their recent equivalents are characterized by pyrolysis-mass spectrometry and pyrolysis-g...

Hydrocarbon phenotyping of algal species using pyrolysis-gas chromatography mass spectrometry

BACKGROUND Biofuels derived from algae biomass and algae lipids might reduce dependence on fossil fuels. Existing analytical techniques need to facilitate rapid characterization of algal species by phenotyping hydrocarbon-related constituents. RESULTS In this study, we compared the hydrocarbon rich algae Botryococcus braunii against the photoautotrophic model algae Chlamydomonas reinhardtii u...

Characterization of exopolymers of aquatic bacteria by pyrolysis-mass spectrometry.

Exopolymers from a diverse collection of marine and freshwater bacteria were characterized by pyrolysis-mass spectrometry (Py-MS). Py-MS provides spectra of pyrolysis fragments that are characteristic of the original material. Analysis of the spectra by multivariate statistical techniques (principal component and canonical variate analysis) separated these exopolymers into distinct groups. P...

On-Probe Pyrolysis Desorption Electrospray Ionization (DESI) Mass Spectrometry for the Analysis of Non-Volatile Pyrolysis Products.

An on-probe pyrolyzer has been constructed and interfaced with Desorption Electrospray Ionization (DESI) Mass Spectrometry (MS) for the rapid analysis of non-volatile pyrolysis products. The detection and analysis of non-volatile pyrolysis products of peptides, proteins and the synthetic polymer poly(ethylene glycol) are demonstrated with this instrument. The on-probe pyrolyzer can be operated ...
