چکیده

Glutamine Synthetase from Bacillus brevis Bb G1was purified from pyruvate grown cells(GSpyr). The effect of ligands on the tryptophan fluorescence of the purified enzyme GSpyrwas investigated. With increasing concentrations of Lglutamine or glycine in GSpyr, a shift in emission maximum, change in fluorescence intensity and change in life times were observed compared to the emission maximum, fluorescence intensity and life times of GSpyr. These observations strongly support the possibility that GSpyr undergoes a conformational change on binding with ligands and each ligand produced differentconformational changes in GSpyr. Also, different concentrations of each ligand produced different protein conformations in theenzymeGSpyr.

Fluorescence study on ligand induced conformational changes of glutamine synthetase from Bacillus brevis Bb G1 under sporulating conditions

Article info: Received: 25 June 2014 Accepted: 13 November 2014 ABSTRACT Glutamine synthetase, an important enzyme of nitrogen metabolism, was purified under sporulating conditions (GSala). The effect of ligands on the tryptophan fluorescence of the purified enzyme GSala was investigated. With increasing concentrations of L-glutamine in GSala, a blue shift in emission maximum with an increase i...

Conformational changes induced by Mg2+ on the multiple forms of glutamine synthetase from Bacillus brevis Bb G1

Article info: Received: 28 June 2013 Accepted: 15 August 2013 ABSTRACT Conformational changes play an important role in the function of proteins. Glutamine synthetase, an important enzyme of nitrogen metabolism, was purified under sporulating (GSala) and non-sporulating (GSpyr) conditions and the effect of Mg on these multiple forms was studied by fluorescence spectroscopy to detect possible co...

COMPARISON OF DIFFERENT MOLECULAR FORMS OF GLUTAMINE SYNTHETASE FROM BACILLUS BREVIS Bb G1 BY FLUORESCENCE SPECTROSCOPY

Glutamine Synthetase in Bacillus brevis Bb G1was purified from alanine grown cells (GSala) and pyruvate grown cells (GSpyr). The emission maximum, life times and quantum yield were found to be 329 nm, 5.8ns & 1.8ns and 0.097 for GSala and 320nm, 5.08ns & 1.3ns and 0.032 for GSpyr respectively. The large wavelength shift in the emission maximum, the significant differences in the shape of the sp...

A fluorescence spectroscopic study of glutaminyl-tRNA synthetase from Escherichia coli and its implications for the enzyme mechanism.

Interaction between Escherichia coli glutaminyl-tRNA synthetase (GlnRS) and its substrates have been studied by fluorescence quenching. In the absence of other substrates, glutamine, tRNA(Gln) and ATP bind with dissociation constants of 460, 0.22 and 180 microM, respectively. The presence of other substrates has either no effect or, at best a weak effect, on binding of ligands. Attempts to isol...

Feedback regulation in the anthranilate aggregate from wild type and mutant strains of Escherichia coli.

The anthranilate aggregate, which catalyzes the first two reactions of tryptophan biosynthesis in Escherichia coli, consists of two anthranilate synthetase subunits and two phosphoribosyltransferase subunits. The aggregate remains associated under physiological conditions. Three lines of evidence indicate that regulation in the aggregate involves conformational changes associated with the bindi...
