چکیده
BACKGROUND/AIM: Recent studies have suggested a relation of homocysteine with lipid metabolism. The aim of this study was to analyze a possible genetic basis for such a relation in 504 individuals including 135 consecutive Caucasian patients diagnosed with cerebrovascular disease as well as the patients’ healthy spouses (n = 100) and offspring (n = 269). METHODS: We analyzed the association of plasma levels of lipoprotein(a), total cholesterol, low-density lipoprotein (LDL), high-density lipoprotein (HDL), and triglycerides with plasma homocysteine levels and with the following 7 variants of homocysteine metabolism: dihydrofolate reductase c.594 + 59del19bp, cystathionine -synthase c.844855ins68, methioninesynthasec.2756AG, methylenetetrahydrofolatereductasec.677CTandc.1298AC, reducedfolatecarrier1c.80GA, andtranscobalamin2(Tc2)c.776CG.RESULTS : LinearregressionanalysisshowedanassociationofTc2c.776CGwithLDL(p = 0.010), HDL(p = 0.009), andTG(p = 0.007), withtheGalleleofTc2c.776CGassociatedwithanunfavorablebloodlipidprofile.Moreover, theGalleleofTc2c.776CGwasassociatedwithhigherhomocysteineplasmalevelsinthesubgroupofpatients(p = 0.013, 1−wayANOV A).CONCLUSION : Thesedatasupportthehypothesisthatalterationsinhomocysteinemetabolismandanunfavorablebloodlipoproteinprofilemayhaveacommongeneticbasis.Suchconditionsmayberelevantforstudiesinvestigatingindependentriskfactorsforvasculardisease.Copyrightľ2010S.KargerAG, Basel. Homocysteine plasma levels in patients treated with antiepileptic drugs depend on folate and vitamin B12 serum levels, but not on genetic variants of homocysteine metabolism.
BACKGROUND Antiepileptic drugs (AEDs) are commonly used in the treatment of epilepsy, psychiatric diseases and pain disorders. Several of these drugs influence blood levels of folate and vitamin B12 and, consequently, homocysteine. This may be relevant for AED effects and side effects. However, not only folate and vitamin B12, but also genetic variants modify homocysteine metabolism. Here, we a...

متن کاملPolymorphisms of homocysteine metabolism are associated with intracranial aneurysms.
BACKGROUND Impaired homocysteine metabolism is associated with a number of vasculopathies including extracranial aneurysms. We analyzed the possible association of nine genetic variants of homocysteine metabolism with the occurrence of intracranial aneurysms. METHODS Caucasian patients (n = 255) treated at two German hospitals for intracranial aneurysms and local controls (n = 348) were genot...
متن کاملModulation of cystathionine beta-synthase level regulates total serum homocysteine in mice.
Elevated total plasma homocysteine is an independent risk factor in the development of vascular disease in humans. Cystathionine beta-synthase (CBS) is an enzyme that condenses homocysteine with serine to form cystathionine. In this article, we describe the effects of modulating CBS activity using a transgenic mouse that contains the human CBS cDNA under control of the zinc-inducible metallothi...
متن کاملHomocysteine metabolism.
Homocysteine is a sulfur amino acid whose metabolism stands at the intersection of two pathways: remethylation to methionine, which requires folate and vitamin B12 (or betaine in an alternative reaction); and transsulfuration to cystathionine, which requires pyridoxal-5'-phosphate. The two pathways are coordinated by S-adenosylmethionine, which acts as an allosteric inhibitor of the methylenete...
