Osteopontin and its Integrin Receptor avb3 Are Upregulated During Formation of the Glial Scar After Focal Stroke

چکیده
Background and Purpose—Microglia and astrocytes in the peri-infarct region are activated in response to focal stroke. A critical function of activated glia is formation of a protective barrier that ultimately forms a new glial-limiting membrane. Osteopontin, a provisional matrix protein expressed during wound healing, is induced after focal stroke. The present study was performed to determine the spatial and temporal expression of osteopontin and its integrin receptor avb3 during formation of the peri-infarct gliotic barrier and subsequent formation of a new glial-limiting membrane. Methods—Spontaneously hypertensive rats (n519) were subjected to permanent occlusion of the middle cerebral artery and killed 3, 6, and 24 hours and 2, 5, and 15 days after occlusion. The spatial and temporal expression of osteopontin mRNA was determined by in situ hybridization, and that of osteopontin ligand and its integrin receptor avb3 was determined by immunohistochemistry. Results—Osteopontin mRNA was expressed de novo in the peri-infarct region from 3 to 48 hours; by 5 days osteopontin mRNA expression was restricted to the infarct. Osteopontin protein was expressed by peri-infarct microglia beginning at 24 hours and by microglia/macrophages at 48 hours in the infarct. Integrin receptor avb3 was expressed in peri-infarct astrocytes at 5 and 15 days. Conclusions—Early microglial/macrophage expression of osteopontin mRNA defines the borders and final infarct area at 24 hours. At 5 days osteopontin ligand is at a distance from the peri-infarct astrocytes expressing integrin receptor avb3. By 15 days astrocytes expressing integrin receptor avb3 are localized in an osteopontin-rich region concomitant with formation of the new glial-limiting membrane. The de novo expression and interaction of osteopontin ligand with its receptor integrin avb3 suggest a role in wound healing after focal stroke. (Stroke. 1998;29:1698-1707.)

Smooth Muscle Cell Matrix Metalloproteinase Production Is Stimulated via avb3 Integrin
This study tests the hypothesis that avb3 integrin receptors play a critical role in smooth muscle cell (SMC) migration after arterial injury and facilitate migration through the upregulation of matrix metalloproteinase (MMP) activity. We showed that b3 integrin mRNA was upregulated by SMCs in the balloon-injured rat carotid artery in coincidence with MMP-1 expression and early SMC migration. T...

To identify kinases that regulate integrin recycling, we have immunoprecipitated avb3 integrin from NIH 3T3 fibroblasts in the presence and absence of primaquine (a drug that inhibits receptor recycling and leads to accumulation of integrins in endosomes) and screened for co-precipitating kinases. Primaquine strongly promoted association of avb3 integrin with PKD1, and fluorescence microscopy i...

Bone sialoprotein (BSP) is a protein thought to be highly specific for bone. BSP contains an arginine-glycineaspartic acid (RGD) cell attachment sequence involved in osteoclast adhesion to bone matrix via the vitronectin receptor and plays an important role in the early process of bone mineralization and resorption. The study by Bellahcène et al1 in this issue of Circulation Research indicates ...
متن کاملOsteopontin Augments M2 Microglia Response and Separates M1- and M2-Polarized Microglial Activation in Permanent Focal Cerebral Ischemia
Background Focal cerebral ischemia induces distinct neuroinflammatory processes. We recently reported the extracellular phosphor-glyco-protein osteopontin (OPN) to directly affect primary microglia in vitro, promoting survival while shifting their inflammatory profile towards a more neutral phenotype. We here assessed the effects of OPN on microglia after stroke in vivo, with focus on infarct d...
متن کاملOsteopontin promotes vascular cell adhesion and spreading and is chemotactic for smooth muscle cells in vitro.
Osteopontin is an Arg-Gly-Asp-containing acidic phosphoprotein recently shown to be upregulated in vascular smooth muscle during rat arterial neointima formation and in human atherosclerotic plaques. Functional studies showed that osteopontin promoted adhesion of both cultured aortic endothelial cells and aortic smooth muscle cells. Adhesion of vascular cells to osteopontin was dose dependent a...
