The effect of inhibitors on photosynthetic electron transport chain in canola leaf discs
نویسندگان
چکیده
The mechanisms of photosynthetic electron transport can be elucidated by inhibition of electron flow through the use of specific substances that, when combined with the chlorophyll chlorophyll a fluorescence emission was measured to investigate the effect of several inhibitors of the photosynthetic electron transport chain in canola leaf discs. Leaf discs were incubated in the dark for 2 hours in different solutions: (a) water – control; (b) DCMU at 500 μM; (c) phenanthroline at 10 mM; and (d) hydroxylamine at 10, 50, 100 and 200 mM. Similar effects were observed between DCMU and phenanthroline, the initial fluorescence value was altered, but not the maximum fluorescence, with the disappearance of the IP phase. Hydroxylamine interacted and inhibited the oxygen evolving complex and caused an imbalance between the rate of QA reduction by photosystem II and the rate of QA oxidation by photosystem I.

متن کاملEthylene formation in sugar beet leaves: evidence for the involvement of 3-hydroxytyramine and phenoloxidase after wounding.
Ethylene production by sugar beet (Beta vulgaris L.) leaf discs is inhibited by white (or red, >610 nm) light or by wounding. In contrast, in wounded leaf discs, ethylene production is stimulated by light. The effect of light on wounded leaf discs has been studied by using an in vitro system which mimics the loss of compartmentation in the wounded leaf. Chlorophyll-free extracts from sugar beet...

The aim of this study is to evaluate the effects of water deficit stress on chlorophyll fluorescence (CF) characteristics of photosystem II (PSII) and pigment contents in two rootstock seedlings (Pistacia atlantica L. and P. khinjuk L.). Three levels of soil water potential (Ψs) was used, including WWD (-0.05 MPa), MWD (-0.7 MPa) and SWD (-1.5 MPa). It was found that water stress increased the...

The photosynthetic phosphoenolpyruvate carboxylase (C(4)-PEPC) is regulated by phosphorylation by a phosphoenolpyruvate carboxylase kinase (PEPC-k). In Digitaria sanguinalis mesophyll protoplasts, this light-mediated transduction cascade principally requires a phosphoinositide-specific phospholipase C (PI-PLC) and a Ca(2+)-dependent step. The present study investigates the cascade components at...

The changes of Chl a fluorescence induction curve in response to different salinity levels in Canola (Barassica napus, L.) plants were investigated. The canola seedlings were exposed to salt stress (100 mM and 200 mM NaCl) for 7 days and Chl a fluorescence induction kinetics was analysed. By using the OJIP-test, various parameters like VJ, area, N, φPo, φEo, ψo, RC/ABS and PIABS were calculated...
