چکیده

To obtain purified H and M antigens suitable as primary standards in the serological diagnosis of histoplasmosis by agar gel double-diffusion tests, H and M reactive components of histoplasmin were fractionated by column chromatography by using Sephadex G-100, Sephadex G-200, and diethylaminoethyl cellulose. Six fractions from diethylaminoethyl cellulose were reactive in agar gel double-diffusion, complement fixation, and capillary precipitin tests. When examined by electrophoresis on acrylamide gel, one M antigen (fraction 2, molecular weight greater than 200,000) and two H antigens (fractions 5 and 6, molecular weight greater than 200,000) each gave essentially a single protein band. In agar gel double-diffusion and complement fixation tests with sera from patients with proven cases of histoplasmosis, blastomycosis, or coccidioidomycosis, these two fractions of H antigen and the one of M antigen reacted only with sera from proven or suspect cases of histoplasmosis and showed reactivity with those sera known to contain only the anti-H or anti-M antibody, respectively. Fraction 2 (M antigen) and fractions 5 and 6 (H antigens) had carbohydrate-to-protein ratios of 1.60, 0.77 and 0.78, respectively. Both antigens contained galactose, glucose, mannose, and hexosamine, with mannose being the predominant sugar. Fraction 2 was characterized by a high proline and glucose content, whereas fractions 5 and 6 contained higher concentrations of galactose, mannose, glycine, and alanine. Each of these products appeared to separate into two active fractions, one of a molecular weight greater than 200,000 and one of a molecular weight less than 35,000. The M antigen component of fraction 2 was still characterized by a high proline content, whereas the H antigen components of fractions 5 and 6 had a high content of glutamic acid, serine, glycine, and proline.

Immunological studies with an m-deficient histoplasmin skin-test antigen.

Previous studies have shown that a single skin test to histoplasmin may induce complement-fixing antibodies or M precipitins (or both) to histoplasmin in histoplasmin-sensitive, but serologically negative, individuals. Ideally a skin-test antigen should be one which detects hypersensitivity without stimulating humoral antibodies. Histoplasmin skin-test antigens presently used contain both H and...

PURIFICATION AND CHARACTERIZATION OF CELL WALLMANNOPRO TEINS OF CANDIDA ALB/CANS USING INTACT CELL METHOD

Virulence of the opportunistic yeast, Candida albieans, involves the interplay of many complex changes including the yeast-hyphae transition, which mainly involves protein changes. Cell wall mannoproteins are found to be the main cause of adherence of C. albieans to epithel ial cells in the first step of an infection process. In the present study, cell wall mannoproteins of intact yeast wer...

Evaluation of histoplasmin and yeast phase antigens derived from a single strain of Histoplasma capsulatum in the complement fixation test.

A complement fixation procedure for the serological diagnos of histoplasmosis in a human patient was attempted first in 1941 by Van Pernis, Benson, and Hollinger. The results were negative with an antigen which consisted of the undiluted filtrate (histoplasmin) from a glucose broth medium inoculated with Hidtoplasma capsulatum. Since that time other types of antigens have been developed by vari...

PURIFICATION AND CHARACTERIZATION OF PROTEIN ANTIGENS ISOLATED FROM MYCOBACTERIUM TUBERCULOSIS (H37Rv STRA IN) AND THEIR EFFECTS ON CELL-MEDIATED IMMUNE RESPONSES IN GUINEA PIGS

Mycobacterium tuberculosis (H37Rv strain) was used in this study. The bacterial cells were disintegrated by sonication. The separation and characterization of the soluble molecules were attempted by various techniques including gel filtration, ion exchange chromatographies and polyacrylamide gel electrophoresis, using SDS and 2ME. Eight protein molecules with molecular weights ranging from...

Effects of a single histoplasmin skin test on the serological diagnosis of histoplasmosis.

Numerous reports have indicated that a single histoplasmin skin test may stimulate humoral antibodies to Histoplasma capsulatum antigens in histoplasmin-hypersensitive individuals. Although these investigations concur that antibody elevations are evoked, they vary in the reported degree of incidence and response induced, and they cast doubt on the interpretation of serological tests in the diag...
