چکیده

Enzymatic oxidation of 3,4-dihydroxyphenyl-L-alanine (L-DOPA) with laccase from Trametes versicolor was investigated. The highest enzyme activity at pH 5.4 and at 25 °C was found. The reaction kinetics and the effect of dissolved oxygen concentration on the reaction rate were evaluated. A mathematical model, comprised of double-substrate Michaelis-Menten kinetics and mass balances for L-DOPA and dissolved oxygen concentrations, was developed in order to describe and predict the process of L-DOPA oxidation. Kinetic parameters,Vm , K m DOPA L and K m O2 were estimated and experimentally verified by a set of experiments with constant additional aeration for different initial concentrations of L-DOPA and dissolved oxygen. A significant increase in reaction rate was established at a higher oxygen concentration in the inlet gas. The developed model was used to investigate the influence of dissolved oxygen concentration on L-DOPA conversion.

Studies on enzymatic oxidation of 3′,4′-dihydroxy-l-phenylalanine to dopachrome using kinetic isotope effect methods

We report the studies on the mechanism of oxidation of 3',4'-dihydroxy-l-phenylalanine (l-DOPA) to neurotoxic dopachrome catalyzed by enzyme horseradish peroxidase (EC 1.11.1.7) using the kinetic (KIE), and solvent (SIE), isotope effect methods. For kinetic studies two specifically deuterated isotopomers: [2',5',6'-2H3]-l-DOPA was synthesized by the acid catalyzed isotopic exchange between nati...

Determination of reactive oxygen species generated in laccase catalyzed oxidation of wood fibers from Chinese fir (Cunninghamia lanceolata) by electron spin resonance spectrometry.

The aim of the present study was to determine whether the radical reaction intermediates--reactive oxygen species (ROS) were formed during the laccase-catalyzed oxidation of wood fibers from Chinese fir (Cunninghamia lanceolata) and to quantify tentatively its production with electron spin resonance (ESR) spectrometry. To investigate the activation pathways triggered by laccase, ESR spin-trappi...

Mechanistic Studies of Laccase/TEMPO Catalyzed Aerobic Oxidation of Alcohols

The reaction parameters for the Laccase/TEMPO system in the aerobic oxidation of aromatic alcohols were investigated. The optimized concentrations of laccase and alcohol were found to be 10 U/ml and 160 mM respectively. Under these conditions the use of 9 mol % TEMPO led to good conversions within 4 h. The optimum temperature for the reaction was found to be above room temperature (around 30°C).

Protection of wood from microorganisms by laccase-catalyzed iodination.

In the present work, Norway spruce wood (Picea abies L.) was reacted with a commercial Trametes versicolor laccase in the presence of potassium iodide salt or the phenolic compounds thymol and isoeugenol to impart an antimicrobial property to the wood surface. In order to assess the efficacy of the wood treatment, a leaching of the iodinated and polymerized wood and two biotests including bacte...

Cryptococcus neoformans laccase catalyses melanin synthesis from both D- and L-DOPA.

The human fungal pathogen Cryptococcus neoformans produces melanin in the presence of various substrates, including the L enantiomer of 3,4-dihydroxyphenylalanine (DOPA). The enzyme laccase catalyses the formation of melanin by oxidizing L-DOPA, initiating a series of presumably spontaneous reactions that ultimately leads to the polymerization of the pigment in the yeast cell wall. There, melan...
