چکیده
AS-3-Ketosteroid isomerase of Pseudomonas testosteroni is a homogeneous and crystalline enzyme that promotes the irreversible conversion of A5-3-ketosteroids to A4-3-ketosteroids (1, 2). This enzyme is one of the most active catalytic proteins known, since each mole (molecular weight 40,800) converts 16.7 x lo6 moles of A5-androstene-3, 17-dione (I) to A4-androstene-3, 17dione (II) per minute at 25” and pH 7.0, when saturated with the substrate.

On roads not taken in the evolution of protein catalysts: antibody steroid isomerases that use an enamine mechanism.
Reactive immunization has emerged as a new tool for the study of biological catalysis. A powerful application resulted in catalytic antibodies that use an enamine mechanism akin to that used by the class I aldolases. With regard to the evolution of enzyme mechanisms, we investigated the utility of an enamine pathway for the allylic rearrangement exemplified by Delta5-3-ketosteroid isomerase (KS...

The absorption spectra of a series of inhibitors bound at the active site of Delta(5)-3-ketosteroid isomerase from Pseudomonas putida were found to exhibit substantial variations in the contributions of the protonated and deprotonated forms. Systematic variation of the inhibitor solution pK(a) combined with a method of quantifying the contributions of each protonation state showed the oxyanion ...
متن کاملNovel steroid-sensing model and characterization of protein interactions based on fluorescence anisotropy decay.
Intramolecular binding of a ligand with an alkyl link, (-CH(2))(3), covalently bound to a residue near the active site of the protein forms a novel steroid-sensing model. A genetically engineered big up tri, open(5)-3-ketosteroid isomerase (KSI) was designed to conjugate uniquely with this ligand at its Cys-86 through the formation of a disulfide bond. The steady-state protein-ligand binding, m...
متن کاملCrystal structure of delta(5)-3-ketosteroid isomerase from Pseudomonas testosteroni in complex with equilenin settles the correct hydrogen bonding scheme for transition state stabilization.
Delta(5)-3-Ketosteroid isomerase from Pseudomonas testosteroni has been intensively studied as a prototype to understand an enzyme-catalyzed allylic isomerization. Asp(38) (pK(a) approximately 4.7) was identified as the general base abstracting the steroid C4beta proton (pK(a) approximately 12.7) to form a dienolate intermediate. A key and common enigmatic issue involved in the proton abstracti...

In this research, the interaction of [CuL(DMF)], [NiL(DMF)] and [VOL(DMF)] (where L = ((E)-4-((2-amino-5-nitrophenylimino)methyl)benzene-1,3-diol)) complexes derived from tridentate Schiff base ligand with bovine serum albumin (BSA) and DNA was investigated via electronic absorption and fluorescence spectroscopy. The Ultraviolet-Visible (UV-Vis) spectra exhibited an isosbestic point for the com...
