Preferential cleavage of S S and C S bonds in electron detachment dissociation and infrared multiphoton dissociation of disulfide-linked peptide anions

چکیده
Disulfide bonds generally show only limited cleavage in positive ion mode collision activated dissociation (CAD). However, it has been emonstrated that a reverse situation exists in negative ion mode in which preferential S S and C S bond cleavage occurs. Here, we show that lectron detachment dissociation (EDD) and infrared multiphoton dissociation (IRMPD) of peptide anions containing disulfide linkages also result n preferential cleavage of S S and C S bonds. Resulting products are mainly radical ions in EDD whereas IRMPD produces even-electron roduct ions, as expected, thereby supporting different disulfide cleavage mechanisms for these two fragmentation processes. We also show that, n EDD, the presence of tryptophan can result in abundant side chain loss (129 Da), which effectively can compete with disulfide bond cleavage. 2007 Elsevier B.V. Does Electron Capture Dissociation Cleave Protein Disulfide Bonds?
Peptide and protein characterization by mass spectrometry (MS) relies on their dissociation in the gas phase into specific fragments whose mass values can be aligned as 'mass ladders' to provide sequence information and to localize possible post-translational modifications. The most common dissociation method involves slow heating of even-electron (M+nH) n+ ions from electrospray ionization by ...
متن کاملInfrared multiphoton dissociation and electron capture dissociation of high-mannose type glycopeptides.
The combination of electron capture dissociation (ECD) and infrared multiphoton dissociation (IRMPD) for the structural characterization of high-mannose type glycopeptides is explored in depth for the first time. Contrary to previous applications to other glycan types, our analyses reveal that IRMPD does not necessarily selectively induce glycan cleavage in high-mannose type glycopeptides; rath...
متن کاملRadical cascades in electron transfer dissociation (ETD) - implications for characterizing peptide disulfide regio-isomers.
Direct characterization of peptides with multiple disulfide bonds by mass spectrometry is highly desirable. In this study, electron transfer dissociation (ETD) of peptide disulfide regio-isomers was studied using model peptides containing two intrachain disulfide bonds. ETD provided rich sequence information (c/z ions) even for the backbone region under the coverage of two disulfide bonds. This...
متن کاملH2 Elimination and C-C Bond Cleavage of Propene: A Theoretical Research
Propene dissociation channels were characterized by ab initio CCSD(T)/6-311++g(d,p) calculations. Inthis work the detailed mechanism of propene dissociation to C2H4+CH2, C2H2+H+CH3, C2H2+CH4 andC3H3+H2+H have been investigated. According to our calculations, ten fragments can be classified intofive dissociated channels. Our results point out that two mechanisms come into play in the H2 eliminat...
متن کاملSO2-* electron transfer ion/ion reactions with disulfide linked polypeptide ions.
Multiply-charged peptide cations comprised of two polypeptide chains (designated A and B) bound via a disulfide linkage have been reacted with SO2-* in an electrodynamic ion trap mass spectrometer. These reactions proceed through both proton transfer (without dissociation) and electron transfer (with and without dissociation). Electron transfer reactions are shown to give rise to cleavage along...
