Limitations on detection of gradients of diffusible chemicals by axons

چکیده
This paper applies a simple analysis of the statistical noise inherent in sensing concentrations of di!usible chemical factors to the problem of guidance of developing axons. We show that growth cones may be able to detect chemical changes of 0.5% across their width, and that guidance by a gradient is limited to distances below about 1 cm, even for a gradient of ideal shape. Axon guidance by diffusible chemoattractants: a gradient of netrin protein in the developing spinal cord.
Gradients of diffusible long-range attractant and repellent proteins have been proposed to guide growing axons during nervous system development, but such gradients have never been visualized directly. In the embryonic spinal cord, commissural axons pioneer a circumferential trajectory to the floor plate at the ventral midline directed by secreted proteins of the netrin family. In the embryonic...
متن کاملSurround Repulsion of Spinal Sensory Axons in Higher Vertebrate Embryos
We have tested whether the orientation of axons sprouting from bipolar dorsal root ganglion neurons is influenced by diffusible cues from surrounding tissues. Surface ectoderm, dermomyotome, and notochord exert strong chemorepulsion on axons growing in collagen gels, operating at separations beyond those found in vivo and active in cocultures of chick and mouse tissues. Basal and alar plates of...
متن کاملOrientation of commissurai axons in vitro in response to a floor plate- derived chemoattractant
Developing axons are guided to their targets by molecular cues in their local environment. Some cues are shortrange, deriving from cells along axonal pathways. There is also increasing evidence for longer-range guidance cues, in the form of gradients of diffusible chemoattractant molecules, which originate from restricted populations of target cells. The guidance of developing commissurai axons...

متن کاملA Mathematical Model of Axon Guidance by Diffusible Factors
In the developing nervous system, gradients of target-derived diffusible factors play an important role in guiding axons to appropriate targets. In this paper, the shape that such a gradient might have is calculated as a function of distance from the target and the time since the start of factor production. Using estimates of the relevant parameter values from the experimental literature, the s...
