Automated Microinjection of Recombinant BCL-X into Mouse Zygotes Enhances Embryo Development

چکیده
Progression of fertilized mammalian oocytes through cleavage, blastocyst formation and implantation depends on successful implementation of the developmental program, which becomes established during oogenesis. The identification of ooplasmic factors, which are responsible for successful embryo development, is thus crucial in designing possible molecular therapies for infertility intervention. However, systematic evaluation of molecular targets has been hampered by the lack of techniques for efficient delivery of molecules into embryos. We have developed an automated robotic microinjection system for delivering cell impermeable compounds into preimplantation embryos with a high post-injection survival rate. In this paper, we report the performance of the system on microinjection of mouse embryos. Furthermore, using this system we provide the first evidence that recombinant BCL-XL (recBCL-XL) protein is effective in preventing early embryo arrest imposed by suboptimal culture environment. We demonstrate that microinjection of recBCL-XL protein into early-stage embryos repairs mitochondrial bioenergetics, prevents reactive oxygen species (ROS) accumulation, and enhances preimplantation embryo development. This approach may lead to a possible treatment option for patients with repeated in vitro fertilization (IVF) failure due to poor embryo quality.

P-26: The Effect of Zygote and 2-cell Development Stages on Vitrification Process of Mouse Embryo
Background: While it is possible to routinely cryopreserve embryos from several mammalian species, the cryopreservation of embryos has largely been limited by their high sensitivity to chilling injury. Many factors such as the stage of embryonic development, cryoprotectant toxicity, the composition of the vitrification solution and cooling and warming rates can influence survival of embryos aft...
متن کاملDetermination the Effects of Epidermal Growth Factor on Developmental Rates of Pre-Implantation Mouse Embryo
Purpose: To determine the effects of different doses of addition of exogenous epidermal growth factor (EGF) on development of pre-implantation mouse embryo. Materials and Methods: Mouse zygotes, two cell, 8 cell any morulae were collected from superovulated NMRI mice 24, 48, 64 and 80 hrs after hCG injection, respectively. The obtained embryos were cultured on medium alone, medium with 1, 4 an...
متن کاملEnhancement of NMRI Mouse Embryo Development In vitro
Most of the systematic studies used in the development of human embryo culture media have been done first on mouse embryos. The general use of NMRI outbred mice is a model for toxicology, teratology and pharmacology. NMRI mouse embryo exhibit the two-cell block in vitro. The objective of this study was to evaluate and compare the effects of four kinds of culture media on the development of zygo...
متن کاملGenomic DNA damage in mouse transgenesis.
Creating transgenic mammals is currently a very inefficient process. In addition to problems with transgene integration and unpredictable expression patterns of the inserted gene, embryo loss occurs at various developmental stages. In the present study, we demonstrate that this loss is due to chromosomal damage. We examined the integrity of chromosomes in embryos produced by microinjection of p...
