چکیده

Two structurally similar bis-phenanthridinium–adenine conjugates (equipped with one or two adenines, respectively), exhibiting strong ds-DNA/RNA groove binding, revealed ratiometric fluorescent recognition of alternating AT-DNA with respect to other ds-DNA/RNA and ss-RNA. Further, CD spectra pattern of adenine–bis-phenanthridinium conjugate/polynucleotide complexes strongly depended on polynucleotide secondary structure. Attached adenine was essential for spectrophotometric recognition response, ds-DNA stabilisation and orientation of compounds regarding chiral DNA helix. © 2015 taylor & Francis

Interactions of acyclic and cyclic bis-phenanthridinium derivatives with ss- and ds-polynucleotidesq,qq

The acyclic bis-phenanthridinium ligands 1, 2 and the cyclic analogue 3 bind to ss-RNA by bis-intercalation. Due to it’s shorter linker 1 exhibits mono-intercalative binding to ds-polynucleotides, while a mixed mode of binding with 2 is shown to strongly dependent on the base composition and tertiary structure of ds-DNA and RNA. The cyclic analogue 3 binds to ds-polynucleotides by non-intercala...

Molecular recognition of AT-DNA sequences by the induced CD pattern of dibenzotetraaza[14]annulene (DBTAA)–adenine derivatives

An investigation of the interactions of two novel and several known DBTAA-adenine conjugates with double-stranded DNA and RNA has revealed the DNA/RNA groove as the dominant binding site, which is in contrast to the majority of previously studied DBTAA analogues (DNA/RNA intercalators). Only DBTAA-propyladenine conjugates revealed the molecular recognition of AT-DNA by an ICD band pattern > 300...

Nucleobase-Guanidiniocarbonyl-Pyrrole Conjugates as Novel Fluorimetric Sensors for Single Stranded RNA.

We demonstrate here for the first time that a guanidiniocarbonyl-pyrrole (GCP) unit can be applied for the fine recognition of single stranded RNA sequences-an intuitively unexpected result since so far binding of the GCP unit to ds-DNA or ds-RNA relied strongly on minor or major groove interactions, as shown in previous work. Two novel nucleobase-GCP isosteric conjugates differing in the flexi...

Recognition of homo-polynucleotides containing adenine by a phenanthridinium bis-uracil conjugate in aqueous media.

Among novel bis-nucleobase-phenanthridinium conjugates bis-uracil analogue stabilized significantly more effective poly-dA-poly-dT and poly-AH(+)-poly-AH(+) than adenine analogue and reference compound . For the alternating poly-dAdT-poly-dAdT however, the binding preference is lost, pointing to the importance of specific interactions of uracils of with homopolynucleotides containing consecutiv...

The size of the aryl linker between two polyaza-cyclophane moieties controls the binding selectivity to ds-RNA vs. ds-DNA.

Aryl-linked (pyridine- vs. phenanthroline-) bis-polyaza pyridinophane scorpiands PYPOD and PHENPOD strongly bind to the double stranded DNA and RNA, whereby very intriguing RNA over DNA selectivity is finely tuned by aryl-linker length and aromatic surface. Moreover, PYPOD and PHENPOD dimer formation at high compound/polynucleotide ratios is highly sensitive to the fine interplay between the st...
