Impact of Retroactivity on Transcriptional Components

چکیده
• Studies show that interconnection of biological parts are vulnerable to Retroactivity, an effect that arises when an upstream system fails to preserve its input/output behavior upon connection to a downstream system. This effect alters both the dynamic as well as the steady state response of the upstream system. Modular cell biology: retroactivity and insulation
Modularity plays a fundamental role in the prediction of the behavior of a system from the behavior of its components, guaranteeing that the properties of individual components do not change upon interconnection. Just as electrical, hydraulic, and other physical systems often do not display modularity, nor do many biochemical systems, and specifically, genetic networks. Here, we study the effec...
متن کاملTHE IMPACT OF RETROACTIVITY ON THE BEHAVIOR OF BIO-MOLECULAR SYSTEMS A Review of Recent Results
Modularity is a powerful property for analyzing the behavior of a system on the basis of the behavior of its components. According to this property, any two components maintain their behavior unchanged upon interconnection. Is modularity a natural property of bio-molecular networks? In this review, we summarize recent theoretical and experimental results that demonstrate that the answer to this...
متن کاملThe Impact of Retroactivity on the Input/output Static Characteristics of a Signaling Component
In prior work, we have shown that just as in many engineering systems, impedance-like effects appear at the interconnection of biomolecular systems. These effects are called retroactivity, to extend the notion of impedance to biological systems. Signaling components, such as covalent modification cycles, play a central role in the transmission of signals within a cell and from outside the cell....
متن کاملThe effect of retroactivity on the transfer function of a phosphorylation system Citation
It was theoretically shown that impedance-like effects, called retroactivity, in biomolecular circuits can significantly impact the behavior of a system. In this paper, we quantify the effect of retroactivity on the transfer function of a phosphorylation system after linearization of its nonlinear model about a steady state. Our analysis shows that retroactivity shifts the poles of the transfer...
متن کاملStochastic modular analysis for gene circuits: interplay among retroactivity, nonlinearity, and stochasticity.
This chapter introduces a computational analysis method for analyzing gene circuit dynamics in terms of modules while taking into account stochasticity, system nonlinearity, and retroactivity. (1) ANALOG ELECTRICAL CIRCUIT REPRESENTATION FOR GENE CIRCUITS: A connection between two gene circuit components is often mediated by a transcription factor (TF) and the connection signal is described by ...
